  • Changes of Root Architecture and Phosphorus Uptake by Roots of Malus hupehensis(Pamp) Rehd. under the Condition of Phosphorus-deficiency

  • To probe into the adaptive mechanism of apple to phosphorus deficiency, a systematic study was made, with Malus hupehensis(Pamp) Rehd adopted as materials and by means of water-culture approach, about the dynamic changes of uptake of phosphorus by roots and root architecture parameters of M. hupehensis R. under the condition of P-deficiency and re-supplied P. Our results revealed that the greatest rate of absorption (Imax) of M. hupehensis R. young seedling in P-deficiency nutrient solution was constantly higher than that of M. hupehensis R. young seedling in full Hoagland solution, and tends to be relatively stable in 120 h; Michaelis constant (Km) went up during 0-12 h, but obviously decreased between 72 to 120 h. In contrast, the effect of root system on the phosphorus transport efficiency in not remarkable, but the phosphorus affinity decreased and gradually returned to normal from 12 h to 120 h when the plants cultivated in P-deficiency solution were transplanted into and stayed in the full Hoagland solution. In addition, the lengths of primary lateral roots in P-deficiency solution were shorter than that of primary lateral roots in full Hoagland solution on the 11th day, and obviously increased from 17th day to 26th day. The results demonstrated that the adaptation of M. hupehensis R. to the condition of phosphorus-deficiency was realized by the changes of parameters of P uptake kinetics during a short time and by the changes of root architecture during a long time.

  • The responses of fruit peel to natural strong sunlight after heat acclimation were studied with bag-removed apples. The results indicated that, the activities of peroxidase (POD) and polyphenol oxidase (PPO) in apple peel increased as a consequence of heat acclimation, and the contents of epicatechin, chlorogenic acid and salicylic acid (SA)increased. Meanwhile, the total antioxidant activities(TAA) of extraction from apple peel increased. Fruits exposed to natural strong sunlight after heat acclimation maintained higher antioxidant ability in fruit peel sometime during explosion, which suggested that preheating acclimation may render a higher ability of scavenging oxygenic free radicals to delay the appearance of fruit sunburn.

  • Ten cultivars with extremely close relationship from Niitaka line pear were used as materials for S-genotyping. In addition, two cultivars 'Imamuraaki' and 'Amanogawa' that were considered to be the parents of cultivar 'Niitaka' were also included. PCR-RFLP analysis of genomic DNAs of the 12 cultivars, nucleotide sequencing of amplified fragments and sequence analysis were performed. As the results, the ten cultivars of Niitaka line were genotyped as follows: 'Niitaka' (S3S9), 'WhangKeumBae' (S3S4), 'Sujeong' (S3S9), 'Josengwhangkeum'(S4S9), 'Soumituniitaka'(S3S9), 'Yewang'(S3S9), 'Jinqiu'(S3S9), 'Banndainiitaka' (S3S9), 'Nokaniitaka'(S3S9), and 'Sunhwang' (S3S5). The S-genotype of 'Amanogawa' and 'Imamuraaki'was identified as S1S9 and S1S6, respectively, suggesting that 'Imamuraaki'is unlikely to be the pollen parent of 'Niitaka'.

  • A Comparative Study on Metabolism of Reactive Oxygen Species during Ovule Development in Seeded And Seedless Grapes

  • The O2- production rate and H2O2 content and the activities of the antioxidant enzymes in the ovule of seedless grape ‘Youngle’and seeded grape ‘Jingxiu’were studied in the different periods after bloom. The results indicated that the concentrations of O2- rose all the time, and the increasing rate in the ovule of seedless cultivar ‘Youngle’was greater than that in the ovule of seeded cultivar ‘Jingxiu’during middle and later periods of development; H2O2 concentration increased initially and decreased before it increased again. The subsequent increase in the seedless cultivar ‘Youngle’was significantly greater than seeded cultivar‘Jingxiu’. The activities of SOD, CAT and AsA-POD increased initially and dropped drastically after ovule abortion in the seedless cultivar‘Youngle’. But it maintained high level in seeded cultivar‘Jingxiu’. The activity of POD in the ovule of the seedless cultivar ‘Youngle’displayed a similar trend to the other enzymes, but the activity in seeded cultivar‘Jingxiu’ increased constantly, but the increasing speed was lower in later stages. The changes of reactive oxygen content and the activity of antioxidant enzymes in ovules have some relevance to ovule abortion in the seedless cultivar 'Youngle'.

  • The changes in Hunter L, a, b or browning index and activities of polyphenoloxidase (PPO) and peroxidase (POD), total phenol content in fresh-cut peach(Prunus persica L.) treated by 4-hexylresorcinol (4-HR) were measured during storage at 0℃and 5℃. The results showed that treatments with 0.01% 4-HR, 0.01% 4-HR+1% citric acid (CA), 0.01% 4-HR+1% ascorbic acid (AA) and 0.01% 4-HR+1%CA+1%AA reduced the decline of Hunter L and the increase of Hunter a or b, to a various extent, delayed the time in flesh browning, inhibited activities of PPO and POD, and maintained total phenol contents. However, inhibitory effect of the browning by 4-HR was not significant.

  • Exogenous hydrogen peroxide, and salicylic acid and methyl jasmonate,which induced the accumulation of endogenous hydrogen peroxide in plants, were used to investigate the role of hydrogen peroxide on induction of chilling resistance of postharvest banana fruits. The results indicated that 20 mmol·L-1 hydrogen peroxide, 0.7 mmol·L-1 salicylic acid and 0.1 mmol·L-1 methyl jasmonate treatments delayed the occurrence of chilling injury symptom of banana fruits by 2-5 d at 7℃. The treatments also delayed the increase of membrane permeability and MDA contents, induced the activities of SOD and POD, inhibited the activities of CAT, and therefore resulted in accumulation of endogenous hydrogen peroxide in pericarp of banana fruits. The results implied that salicylic acid and methyl jasmonate might induce the accumulation of endogenous hydrogen peroxide by inhibition of CAT activity, and hydrogen peroxide might involve in induction of the chilling resistance of postharvest banana fruits by acting as the signal molecule.

  • The thick liquid disease fruit of dongzao jujube is one of the most important diseases in main producing areas in Shandong Province. The isolation and pathogenicity tests showed that the disease was caused by Macrophomina phaseolina. And the biological characteristics of M. phaseolina were studied in this paper. The lab testing results showed that the median of inhibitory concentration(EC50) of Ovraclostrobin 25%EC, Ovraclostrobin-metiram 60%WG, Azoxystrobin 25%SC and Kresoxim-methyl 50%DF for M. phaseolina were 0.1970, 0.5770, 4.4042 and 55.3120 μg/mL, respectively. The field trial results demonstrated that the control efficacy of 330 mg/L Kresoxim-methyl 50%DF, 250 mg/L Ovraclostrobin 25%EC or 500 mg/L Ovraclostrobin-metiram 60%WG against M. phaseolina was 88.51%-97.60%.

  • In the field, the uniform design was used to test the index of Interference of Population Control (IIPC) of alcohol extract of Artocarpus heteropyhllus against the natural population of diamondback moth Plutella xylostella (L.) by different concentrations and span of times. Through assaying and returning of quadratic equation, a model depicting the correlation between IIPC and two factors (A and B) using uniform design was shown. The model equation is Y=0.4628-10.6164A-0.0212B+117.1290A2-0.7762AB(Y-IIPC, A-concentration, B-times). Actually, This model equation be used to check IIPC under the nature condition, the result showed that the composing of 0.05 g·mL-1×3 times gave the best effect of influencing, IIPC was 0.0448, followed by the composing of 0.04 g·mL-1×3 times, IIPC was 0.0688. The alcohol extracts of Artocarpus. heterophyllus was isolated by immersing five kind organic solvates, and that corresponding extracts have separated out. The extracts of first isolating were surveyed about the bioassay. The result showed that ethyl acetate extract of the alcohol extracts of A. heterophyllus had marked effect of oviposition deterrent and antifeedant on diamondback moth, and their rate was 59.16% and 57.34%. Twenty fractions were obtained using silica gel column from the ethyl acetate extract of the alcohol extracts of A. heterophyllus. By the bioassay, the fractions of L1 and L13 and L18 and L19 showed strong effect of antifeeding, their rate of antifeeding was 85.71%, 73.33%, 78.57%, 88.87%, respectively.

  • Pollen submicroscopic morphology of 8 wild species with potential breeding capabilities from Sect. Idaeobatus and Sect. Malachobatus and 17 introduced cultivars of bramble (Rubus L.) from raspberry and blackberry were examined with scanning electron microscopy. The results showed that the pollen grains were prolate and perprolate, 25.52 μm × 15.37 μm - 50.06 μm × 25.65 μm in size. The exine ornamentation was diverse, distinctly different among species even cultivars. For example, it was mainly striate-perforate in Sect. Idaeobatus as well as raspberry cultivars, striate and striate-perforate in blackberry cultivars, which were clearly different from that of any one of 3 species in Sect. Malachobatus. Cluster analysis results indicated that the size and exine ornamentation characters of pollen could reflect the traditional taxonomy of Rubus. A close blood relationship between those raspberry cultivars and the wild species R. coreanus or R. niveus in Sect. Idaeobatus and far blood relationship between those blackberry cultivars and those 3 wild species in Sect. Malachobatus were found.

  • Effect of Low Temperature Stress on Activities of SOD and Enzymes of Ascorbate-Glutathione Cycle

  • To investigate the effect of low temperature stress on ascorbate content , activities of its relative oxidative enzymes and superoxide dismutase (SOD) , the ascorbate and dehydroascorbate content and activities of SOD and four enzymes involved in the ascorbate-glutathione cycle (AsA-GSH cycle) in strawberry were studied. The results showed that under low temperature stress the activities of SOD, ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDAR) increased, while the acitivities of dehydroascorbate reductase (DHAR) and glutathione reductase (GR) decreased. Meanwhile, low temperature stress induced decrease in ascorbate (AsA) content and increase in dehydroascorbate (DHA) content. When low temperature stress continued, more reactive oxygen species (ROS) were produced and could not be removed efficiently by defense system. The results also indicated that APX had a strong ability to oxidize AsA and MDAR was a main enzyme to regenerate AsA in AsA-GSH cycle.

  • Fragaria×ananassa Duch. 'Toyonoka'and Fragaria×ananassa Duch. 'Rosa' were used to study the changes of aroma components and their relative content by using GC-MS, at the same time, alcohol acyltransferase activity was determined with the method of DTNB colorimetry. The majority of aroma components were determined to be esters(32.18%),aldehydes(19.55%) and ketones(16.82%) in ripe Toyonoka. While the majority of aroma components were determined to be aldehydes(41.51%) and ester(23.25%) in ripe Rosa fruit. In the process of strawberry fruit development the relative content of ketones and esters increased, while the relative content of alcohols and aldehydes decreased. In Toyonoka fruit, the relative content of esters was 4 times more at the red ripe stage than those at the pink stage. While the relative content of alcohols decreased from 54.35% at the red ripe stage to 5.17% at the pink stage. The AAT activity increased obviously in Toyonoka and Rosa in the development of the fruit. In addition, the AAT activity was greater in Toyonoka than that in Rosa fruit at various development stages.

  • Effects of NaCl Stress on Photosynthetic Characteristics of Three Cucumber Rootstock Seedlings

  • The effects of NaCl stress (0, 50, 100 and 150 mmol·L-1) on the growth, photosynthesis and chlorophyll fluorescence parameters of three cucumber rootstock seedlings (Cucurbita moschata Duch. 'Chaoji Quanwang', Cucurbita ficifolia 'Heizi Nangua', Lagenaria siceraria Standl. var. clavata Makino 'Chaofeng 8848') were investigated. The results showed that the dry and fresh mass of shoot, plant height, leaf number, net photosynthetic rate, stomatal conductance and transpiration rate of three cucumber rootstock seedlings decreased gradually with increasing of NaCl concentrations. The initial fluorescence yield (Fo) of 'Chaoji Quanwang' and the photochemical quenching (qP) of 'Heizi Nangua' decreased significantly, whereas qP of 'Chaofeng 8848' increased significantly under higher salt stress (150 mmol·L-1 NaCl). The maximal photochemical efficiency of PSⅡ (Fv/Fm) did not decrease and there was no significant difference in the quantum efficiency of PSⅡ photochemistry (ΦPSⅡ) under NaCl stress. The chlorophyll a/b ratio of 'Chaoji Quanwang' and 'Heizi Nangua' seedlings did not change significantly under NaCl salt stress, whereas that of 'Chaofeng 8848' decreased significantly.

  • In order to carrying on a digital, normalize, quantitative and undestroied method for plant disease discriminating, two kinds of familiar cucumber diseases (cucumber anthracnose and cucumber brown speck) were researched in the method of picture processing. Color characteristic parameters coming from different periods of disease simple were used to classification and identification . Using stepwise discriminant analysis to select significant parameters, and the bavesian classifier method was proposed to classify cucumber anthracnose, cucumber brown spot and healthy region. The result shows that the correct discrimination of cucumber anthracnose, cucumber brown spot and healthy region were 96.67%, 93.33% and 100%, respectively. For the test set,the correct discrimination of cucumber anthracnose, cucumber brown spot and healthy region were 83.33%, 80.00% and 100%, respectively. The results show that it is feasible to identify and classify plant diseases using color image statistical characteristics.

  • Relationship Between NAD Kinase and Active Oxygen During Ripening and Senescence of Postharvested Tomato Fruit

  • Tomato (Lycopersicon esculentum Mill.) pericarp at pink stage was used to investigate the changes of NAD kinase (NADK) activity and reactive oxygen during ripening and senescence of postharvest tomato fruit at 4℃ and 20℃. Contents of NAD(H), NADP(H) and hydrogen peroxide (H2O2) and superoxide production rate were determined. The degree of the oxidative damage to the membrane lipids (MDA content) and ripening index (lycopene content) were also examined. The results showed that NADK activity increased sharply at early stage and declined significantly at late stage during ripening and senescence at 4℃, but its activity kept lower at 20℃ except higher activity was observed on the 5th day of storage at 20℃. The change model of NAD(H) content contrasted with one of NADK activity. Higher NADP(H)content was found at early store stage than at late store stage and at 4℃ than at 20℃. Significant increase of lycopene content was observed at late store stage at 20℃, but little change was found at the same stage at 4℃. The maximum MDA content appeared earlier at 20℃ than at 4℃. NADK might be involved in postponing ripening and senescence of postharvest tomato fruit by regulating the content and proportion of NAD(H) to NADP(H) which influenced the O2- production rate and H2O2 content.

  • Tomato Dry Matter Production and Distribution on Different Crops in Solar Greenhouse

  • In this study, a source-sink growth unit, which composed of a fruit truss, three pieces of leaves just below this truss and the corresponding stem, was adopted for understanding the dry matter production of tomato crop in three different planting time: over-winter cultivation planted in Oct. 26 (OW); early-spring cultivation in Feb. 17 (ES) and spring-summer cultivation in Mar. 29 (SS), respectively. The results showed that within a source-sink growth unit, fruit dry matter distribution was between 42.6% and 98.6%. Fruit dry matter distribution decreased by 6% with the increasing of the fruit truss number (the lowest fruit truss is the first truss), supposing the seven fruit trusses simultaneously existed in the same plant. When ripe fruits were picked, the distribution of immature fruit dry matter was increased due to the increase of the fruit sink strength. Dry matter production in different cultivation varied a lot due to the obviously different greenhouse climate. The ratio of fruit dry matter to shoot dry production were 72%、62% and 59% for OW, ES and SS cultivation, respectively. Fruit dry matter distribution of SS cultivation was lower than the other two treatments due to the lower rate of fruit-set, when facing high temperature in summer season. The accumulated dry weight in first 100 days after planting was only 1/3 of that in the later growing 100 days in OW cultivation, which was related to low temperature and low light intensity.

  • This paper studies the megasporogenesis, microsporogenesis and development of female and male megasporogenesis of Capsicum annuum L. by means of routine paraffin-embedded sectioning technique. The results are as follows: Five anthers, four locule. The anther wall comprises an epidermis, endothecium, 2 or 3 middle layers and glandular heterotypic tapetum. Towards the outer side the tapetum is derived from the primary parietal layer, whereas on the inner side it develops from the cells of connective. Both of the two kinds of tapetum contribute to the development of male gametophyte. Archesporium in the stamen is multicellular. There are two rows of microspore mother cells on the transverse section of each locule. Meiotic divisions in microspore mother cells are followed by simultaneous cytokinesis and result in tetrahedral and decussate tetrads. Pollen grains are three-porate and two-celled at mature stage. Free-central placentation and polyovulatus. The ovule is anatropous, unitegmic and tenuinucellal with the endothelium. The single archesporial cell functions directly as megaspore mother cell. The later undergoes meiotic divisions and develops into linear tetrad. The third megaspore is the functional one which undergoes three times mitosis and develops into a mature embryo sac with 7 cells and 7 nuclei. Embryo sac is of Polygonum type. The development of stamen is earlier than that of pistil, they synchronize before blossom. When blossom, the dispersed pollen grains arrive at the stigma of its own pistil so that self-pollination is completed. Here we discuss the derivation of heterotypic tapetum, characteristics of its morpha and structure, and relationship between the tapetum and male sterlity.

  • To investigate the early up-regulated expression gene profile in pepper line HAD149 induced by root-knot nematode (RKN) Meloidogyne incognita, a forward subtracted cDNA library of pepper seedling root tips was constructed using suppression subtractive hybridization (SSH). The cDNA of pepper seedling root tips inoculated with J2 RKN on 12, 24 and 36 h were used as tester, and that from untreated root tips as driver. Totally 309 SSH cDNA fragments were selected with DIG Nonradioactive Nucleic Acid Labeling and Detection System. Through the DNA sequencing, we got 259 ESTs. With the Blast, 30 EST fragments didn't find homologous gene, 229 had homologous genes, and among of them, 71 fragments were function unknown. Base on the observing with microscope and the ESTs function analyzing, we putative that disease resistant genes encode nucleotide-binding /leucine-rich repeat (NBS-LRR) have a key rule to recognizes root knot nematode, result in the signal transduction-related genes expressed, activate a suitable defensive response and protection mechanism, it often including hypersensitive response (HR) and Systemic Acquired Resistance (SAR). In addition, all of these responses are base on the first and secondary metabolites.

  • In order to study the relationship and identify different chromosomes between the A and C genome of Brassica, the labeled genomic DNAs of Chinese cabbage and cabbage were respectively hybridized to their metaphase chromosomes. Nonuniform distribution of the fluorescent labeled probe DNAs were observed on all the chromosomes of two species that were tested. The signal patterns varied between species and were related to the genome DNA probes. When Chinese cabbage genomic DNA as probe, the signals were almost only in the pericentromeric regions and the nucleolus organizer regions(NORs) of Chinese cabbage and cabbage, but the signal regions were larger in former than latter. While cabbage genomic DNA as probe, the signals dispersed along the chromosome lengths of cabbage and enhanced signal bands were shown in all pericentromeres and the NORs; Only were observed in the pericentromeres and the NORs of Chinese cabbage chromosomes, and that all the signal intensity was weaker than with genomic DNA probes of Chinese cabbage. Karyotypes of Chinese cabbage and cabbage were analyzed based on GISH signals. The results provided a molecular cytological basic for identification of interspecific hybrids, their chromosomic components and homologous between A and C genomes.

  • Effects of Covering with Colored Plastic Films on the Pigment Content and Photosynthesis in Ginger Leaves

  • The content of chlorophyll and carotenoid in ginger leaves which were covered with green film, red film and blue film at seedling stage respectively was remarkably higher than that of the control in the whole growth period, but the effects of different treatments were different at different growth stages. At seedling stage, the sequence of the content of chlorophyll and carotenoid in ginger leaves from high to low was blue film, green film and red film treatments in turn, but there was no significant difference between blue film and green film treatments under condition of covering with colored plastic films, while with the ginger growing at vigorous growth stage that treated by green film became gradually higher, blue film in the second and red film was lower after film removal. In the whole growth period, the photosynthetic rate (Pn) of ginger leaves covered with films was all significantly higher than that of control, in which the Pn of ginger leaves treated by the green film was higher, followed by the red film and blue film, which increased by 28.0%, 16.3% and 8.9% respectively compared with the Pn of the control at 11:00 on July 19 during covering with films and increased by 21.3%, 12.5% and 5.9% on September 8 after films removal. Furthermore, covering with colored plastic films could create more suitable environmental conditions, alleviate the photosynthetic depression at noon without changing the diurnal variation of photosynthesis.

  • Biocontrol of Zucchini Powdery Mildew by Endophytic Actinomycetes

  • Fifty strains of endophytic actinomycetes were screened in greenhouse pot experiments and leaf-disc test in lab. Two strains showed stable control effect on zucchini powdery mildew. In the leaf-disc test, the application of cell free filtrate of strains GKSHJA and PR1-8 showed 60.98% and 63.22% disease control efficiency, respectively. The control effect of strain GKSHJA with application 24 h before and after inoculation were 55.22% and 16.44%, respectively. The control effect of strain PR1-8 were 51.94% and 24.39% in the greenhouse potted seedlings inoculated with powdery mildew. The preinoculation application of cell free filtrate showed much higher disease control efficiency than postinoculation application. The applications of 5 times diluted cell free filtrates showed 27.81% (in GKSHJA treatment) and 30.19% (in PR1-8 treatment) less disease index than that in untreatment control. In field experiment, the control effect of biocontrol strains GKSHJA and PR1-8 were 56.33% and 69.88%, respectively, after application of 5 times dilution of cell free filtrate on naturally infected zucchini plants. These results showed that strains GKSHJA and PR1-8 were really potential biocontrol endophytic actinomycetes to control cucurbit powdery mildew and this research made a prior period research foundation for development and application of bio-pesticides on cucurbits powdery mildew.

  • Two hybrids, 'Brunello'× 'Yellow Jiant' and 'Prato'×Lilium pumilum, were identified by karyotype analysis and SRAP techniques. Genetic differences and similarity were compared between the parents and the progenies. The results showed that the two hybrids were both triploids, and contain abundant trabant. Four pairs of SRAP primer with higher polymorphism were used and the hybrid bands and complementary bands were found in the hybrids. Pairwise genetic distances between the hybrids and their female parents are smaller than that between the progenies and their male parents. The results of SRAP were in consistent with that of karyotype analysis. The two hybrids were both true progeny of their parents, and both with a genetic tendency to their female parents.

  • This paper studied the senescence process and the relationship of the membrane-lipid peroxidation with the vase life of cut peony flowers. Six cultivars, including 'Chunxiao', 'Shanhehong', 'Qihua Zhenglu', 'Zifeng Chaoyang', 'Linghua Chenyu' and 'Lanhai Bibo' were investigated through observing the changes of morphologic characteristics and determining the indexes of membrane permeability, MDA content, superoxide anion accumulation rate, SOD activity, CAT activity. The results showed that 'Chunxiao', 'Qihua Zhenglu' and 'Linghua Chenyu' exhibited 3 days' full opening period and could be in full blooming with 8 or 7 days' vase life. 'Shanhehong', 'Zifeng Chaoyang' and 'Lanhai Bibo' exhibited 2 days' full opening period and 6 days' vase life. During the vase-holding, the membrane permeability, MDA contents, superoxide anion accumulation rate increased, and the increasing of them was positive to each other. The SOD and CAT activities increased at first and then declined. The damage of cell membrane because of the membrane-lipid peroxidation might be the causal factor of cut peony flowers senescence.

  • The single and interactive effects of nitrogen supply and light conditions on Hosta 'Blue Umbrella' growth and photosynthetic characteristics were studied in this paper. The Hosta was cultivated with sands under four kinds of light conditions (100%,50%,30% and 10% of natural sunlight respectively) and four kinds of nitrogen supply (0,0.5,1.5 and 2.5 g·kg-1 soil respectively).Results showed that biomass, growth characters, photosynthetic rate and soluble protein content increased at first and then decreased as light density decreased in the same nitrogen supply, the suitable light intensities for Hosta 'Blue Umbrella' were from 30% to 50% of natural sunlight. However, chlorophyll contents increased as light density decreased in the same nitrogen supply. In the different nitrogen supply, biomass, growth characters, photosynthetic characteristic, chlorophyll and soluble protein content increased firstly and then decreased as nitrogen supply increased, the suitable nitrogen supply for Hosta 'Blue Umbrella' was 0.5 g·kg-1.

  • The seeds of Impatiens balsamina L. were treated by space flight, and then heritable mutant plants with floral reversion occurring only in apical flower were obtained in the descendant generation. The mutants kept the basical floral structures, however, showed thicker green petals with serrate margin to some extent. Their stamens became petaloid, prematured even diminished. Histological investigation revealed that chloroplast and palisade tissue appeared in the leaf-like petals. Statistical analysis suggested that significant difference existed between the leaf-like petals and normal petals.

  • Cytological investigation of the triploid bermudagrass showed chromosome number varies between 27 and 30 in somatic cell indicating that some of the plants were aneuploid. In order to study the chromosome variation, the digoxigenin-11-dUTP labeled 45S rDNA was used. Three distictive signals were locating to the triploid bermudagrass prometaphase chromosome centromeres by fluorescence in situ hybridization (FISH). The intensity of three signals was different, and two signals of them showed lines sometimes. The chromosome with the weakest signal was easy to snap, and aneuploid appeared.

  • The changes of carbohydrates distribution in Phalaenopsis' leaves and axillary buds were studied by two different temperature treatments. The experimental group was cultivated at 25℃/20℃(day/night), while the control group (control) was carried out in the plastic greenhouse. The results showed that areas of the young leaves increased faster in control than those of the experimental groups in primary stage of the experiment, but their area of mature leaves were approximately equal; while the dry mass of specific leaf area in experimental group exceeded that of control after 15 d treatment. Reducing sugar content of Phalaenopsis' leaves in experimental group dropped after reaching a peak at 20 d, and then elevated rapidly after 30 d and got the largest value which was 31.08 mg·g-1DM at 50 d. While reducing sugar content in the axillary buds descended gradually. The content of soluble sugar, starch and total sugar in Phalaenopsis' leaves rose under the two treatments, but these carbohydrates of Phalaenopsis' axillary buds in experimental group increased rapidly in 15 d and then decreased slowly, at the same time these substances in axillary buds of control increased during the whole experiment. However, the total carbohydrates contents in both leaves and axillary buds in the experimental group were higher than those in control during the period of treatment.

  • The ribosomal genes (45S rDNA) encoding ribosomal RNA synthesis were located in nuclear organizer region (NOR) in plants.The number of NOR should be corresponding to the number of chromosomes with satellite. The 45S rDNA was probed to the metaphase chromosomes and interphase nucleus from male and female Ginkgo plants using fluorescent in situ hybridization (FISH). The metaphase chromosomes and interphase nucleus of female plants displayed four hybridization signals while male plants displayed three signals, indicating that the female plants had four chromosomes with satellite while the male plants had three chromosomes with satellite only. The sex determination of Ginkgo plants was related likely with the number of chromosomes with satellite.

  • To study the relation between molecular structure and storage potential of pear fruit, the 'Pingguoli' and 'Niitaka' species that have different storage potential, were used as materials for comparing the size change in pear soluble pectin and hemicellulose during storage stage using Ultrogel AcA 22 gel filtration profile method. The results showed that the hemicellulose size of 'Pingguoli' significantly bigger than that of 'Niitaka'. The soluble pectin molecular size in 'Pingguoli' not was significantly different in during storage stage, but the molecular size of hemicellulose significantly changed smaller. Instead, the soluble pectin molecular size of 'Niitaka' significantly changed smaller, at the same time without changes in the molecular size of hemicellulose. So the pears storage potentail was related to the changes of molecular structure of soluble pectin and molecular size of hemicellulose, without direct relation to the molecular structure of hemicellulose.

  • To identify molecular markers linked to pcDw gene determining the dwarf character of pear tree, a population of 111 individuals from the cross of 'Aihuali'(Pyrus communis L.)×'Chili'(Pyrus bretschneideri Rehd.) was tested using 412 random primers by the way of bulked segregate analysis(BSA). A RAPD marker, S1172-940, linked to pcDw gene at a distance of 8.3 cM, was detected. This RAPD marker was converted into a SCAR (Sequence Characterized Amplified Region) marker, SCAR-940, which would provide an effective tool for marker assisted selection for the dwarf trait.

  • The present experiment, with the sweet cherry (Prunus avium L.) 'Hongdeng' as its cultivar, investigated the dynamic changes of the fruit growth and development as well as the endogenous hormones of the pulp. The results showed that: The content of ZRs in the pulp had the highest values in the beginning of the first fast-developed period (5th day after full bloom), and then had a tendency of decrease with the fruit, grew and developed until maturated.The contents of IAA and GAs in the pulp of young fruit were high in the first fast-developed period(5th -15th day after full bloom), then decreased with its development until the end of the core-harden period (25th day after full bloom), and they all had a tendency of increase in the second fast-developed period (25th -40th day after full bloom) until maturity, corresponding with the fruit growth. The content of ABA had the peak values in the 5th, 15th, 35th day after full bloom, similar to the fruit (flower) dropping periods.

  • Studies of Indirect Somatic Embryogenesis of Papaya

  • In order to establish genetic transformation system of foreign genes on small fruit type of papaya, indirect somatic embryogenesis of immature embryos (90-120 days after pollination) of Carica papaya L. 'Meizhonghong' was studied by histological section and observation with a scanning electron microscope. The results showed that the calli were originated from the epidemic cells of the radicle after 8-12 days of culture. After 26 days of culture, the whole embryonary axis turned into calli which differentiated to translucent and mucous, and dry types. The cross-section results showed the calli consisted of two distinct sectors, a loose friable tissue of outer sectors and compact aggregates of inter sectors. Yellow pre-embryonic complexes were derived from calli of the plumula after 38 days of culture. It was observed that most of morphological somatic embryos were normally developed.

  • Effects of soybean-cucumber and wheat-cucumber rotation on soil microbial community DNA sequence diversity were evaluated using random amplified polymorphic DNA (RAPD) method. The results showed that grain-cucumber rotation significantly increased the diversity index, richness index and evenness index of soil microbial community, and the soybean-cucumber rotation was more effective than wheat-cucumber rotation in term of the microbial community. Soil microbial community DNA sequence similarity coefficient between the two rotation soils was 0.5678; However the similarity coefficients between the two rotation soils and control soil were 0.3465 and 0.3124, respectively. Cucumber yields in two rotation soils were also higher than that in control soil significantly. This may imply that rotation can improve soil microbial ecology.

  • Effects of different exogenous Spermidine (Spd) concentration on Polyamines contents , antioxidant enzyme activities, O2ˉ production rate, H2O2 and MDA contents in roots and leaves of cucumber (Cucumis sativus L.) cultivar 'zhongnong 8' in hydroponics were investigated. The results showed that exogenous Spd at 0.05 mmol·L-1 significantly enhanced growth of cucumber seedlings and alleviated the damage of hypoxia stress efficiently. Under hypoxia stress, activities of antioxidant enzymes were associated closely with polyamines contents. The addition of 0.05 mmol·L-1 exogenous Spd obviously increased endogenous leaves of Spd, Spermine (Spm), and (Spd+Spm)/Put ratio, while decreased putrescine (Put) content, enhanced the activities of antioxidant enzymes, While decreased the O2ˉ production rate, H2O2 and MDA contents. Therefore, exogenous Spd at a suitable concentration promoted the growth of cucumber seedlings under hypoxia stress, and improved tolerance of cucumber seedlings to hypoxia stress.

  • Effects of Light Intensity on Photosynthetic Characteristics and Quality of Taraxacum mongolicum

  • Under different light intensity, chlorophyll content, net photosynthetic rate, and quality of Taraxacum mongolicum were studied. The results indicated that with the decrease of light intensity, photosynthetic midday-depress was weakened, contents of crude-fibre and chlorogenic acid were reduced, while contents of chlorophyll and nitrate were increased. Under the 75% natural light intensity (910 μmol·m-2·s-1), the Pn, contents of soluble protein, soluble sugar and vitamin C were the highest among the four treatments; the chlorogenic acid content was higher, contents of nitrate and crude-fibre were lower than the others. When the light intensity for cultivating Taraxacum mongolicum was 910 μmol·m-2·s-1, its photosynthetic efficiency and quality could be improved.

  • The petal, filament and ovule of Clivia miniata Regei 'Youjiang' were used as explants for in vitro culture. The results showed that the explants disinfected with 0.1% HgCl2 for 10 min were most effective with the average contamination rate of 9.9%.The rates of induction and differentiation of three kinds of floral organ explants were different. The petals had the highest induction and differentiation rates of 15.6% and 57.1%, respectively. There was direct relationship between percentage of induction and differentiation of petals and size of flower buds, concentration of hormone and sucrose. The rates of callus induction and differentiation were the highest when the length of flower bud was 0.6-1.0 cm. The optimum medium was MS+2,4-D 2.0 mg·L-1+BA 1.0 mg·L-1+NAA 0.5 mg·L-1+3.0% sucrose. The rate of induction and differentiation of floral organ explants was much higher while the concentration of sucrose was 3%.

  • Thirty-eight species of Dendrobium were analyzed phylogenetically using AFLP technique. Eight pairs of primers combination were screened from 64 pairs of primers combination and generated 1 644 amplified loci, 1 639 of which were polymorphic loci which accounted for 99.7% in the total amplified loci. It showed that the generation diversity standard of Dendrobium was very high in our country. Cluster analysis showed that Sect. Dendrobium and Sect. Formosae did not form an independent branch, Sect. Aporum and Sect. Strongyle were consisted of a common branch. They had a lower similarity with another section. Sect. Grastidium, Sect. Stachyobium and Sect. Stuposa formed an independent branch. They had a different evolution direction. Genetic relationship in domestic Dendrobium was much more complicated, and they were not a monophyletic group in nature, but a paraphyletic or polyphyletic group.

  • The plantlet regeneration, with multi-shoots especially, derived from seed, cotyledon, squama and tip-bud of Cycas revolute Thunb was studied in vitro. The results showed that the germination of seed cultured in vitro could be six months earlier than that of sowing in normal condition. When inoculated on ER+BA 2 mg/L+IBA 0.4 mg/L+juice of Cycas revolute Thunb episperm 2 ml/L,the rate of multi-shoots was 76.7%. The differentiation rate of bud derived from cotyledon and squama was 53.3% and 33.3% , respectively. It will be better when plantlets were transplanted from April to July; and the survival rate was more than 90%.

  • Peach (Prunus persica L.‘Okubao’)fruit was adopted in this experiment. Comparing different effects of two-dimensional electrophoresis (2-DE)gels that were involved with quantitative loading of sample, sample buffer, equilibration buffer and the type of staining to explore influence factors of 2-DE patterns on peach fruit. Although the two-dimensional gel stained by coomassie brilliant blue had gained less protein spots than the gel stained by silver, it had a clear background intensity and was applicable to the proteomic study. Furthermore, approximately 100 μg of protein was brought to a final volume of 375 μL sample buffer which contained thiourea and sulfphobetaine 3-10 and loaded on 17 cm immobilized pH gradients strip, then the strip was incubated in equilibration buffer with tributyl phosphine after isoelectric focusing. The two-dimensional gel stained by silver detected 1 209 protein spots and had slight streaks. The results demonstrated that the protocol was suited to proteomic analysis of peach fruit and compatibled with protein identification by mass spectrometry of downstream processing.

  • In order to establish the technology for separating and purifying total flavonoids from Toona sinensis leaves with macroporous resin, 6 types of macroporous resins were selected to be investigated. The results showed that LSA-10 was the best for separating and purifying total flavonoids in the following conditions: the loaded concentration of total flavones 0.653 mg/mL, pH4.0-5.5, the flow rate 240 mL/h; amount of saturated adsorption for total flavones 440-480mL; 400 mL 50% ethanol as appropriate the eluting reagent, eluting rate 80 mL /h. The purity of total flavonoids purified by LSA-10 was 75.2% when purified four times under the optimal conditions.

  • 'Jin Ai' apricot is an entire mutation of 'Golden Sun' apricot. It has a small tree size and short internodes. The fruit is round with an average mass of 56.9 g. The flesh is yellow and close-textured. The content of soluble solids is 14%. It has abundant juice and tastes tart- sweet. It is free stone and the kernel is bitter. It has a high rate of self-fertility and consecutive high yield. It's suitable for dense-planting culture under protection and for ornamental miniature garden.

  • ‘Caomei Li’ is an excellent litchi cultivar which was selected from litchi seedling in Guangxi.It has good characteristics such as high yield and good quality,the average quality of simple fruit is 27.5 g, the esculent rate is 77.94%,and the aborted-seeded rate is 97.0%. The flesh is waxen yellow with sweet and aroma, the soluble solids content is 17.73%.

  • A new mid-early maturing tomato hybrid,'Zheza 205' has been released. It can be grown normally in low-temperature and poor-beam of light. It is high resistant to ToMV, fusarium wilt,verticillium wilt and mid-resistant to leaf mold. Middle to large fruit, bright red color, regular shape and good commodity with high yield. The yield can be 150-200 t/hm2. Fruit is storable. Its shelf life lasts 25 days in usual temperature (25℃). The variety is suitable for protected production.

  • 'Ziyan 1' is a middle pungent F1 hybrid. It was developed by crossing the domestic good inbred line with the import good inbred line which selected from a variety of purple sweet pepper. It is tolerant to low temperature and weak light and can be harvested for purple fruits in 42 days after transplanting. The yield is 39 000-45 000 kg/hm2. The fruit is ox-horn shaped with purple skin and deep red skin when matured.
