  • 7.2℃ 、0—7.2℃ and Utah model were used as estimation of the chilling requirement of mole than 450 peach varieties at Zhengzhou in Henan from 1986 to 2001.The results showed that: The date of average temperature in autumn equal to or lower than 7.2℃ was appropriate for the initial date of efective chilling accunlLi—lation,0—7.2℃ mod el could be suitable for predicating efectively rest completion;Utah model performed aCELI—
    mtely in normal and long chilling requ irement; but 7.2℃ mod el Was inefective. The correlation coeficient of chilling requirement with leaf bud breaking an d initial blooming, were 0.52 an d 0.58 respectively, both of them reached significant levels.Reference pe ach cuhivam for chining requ irement determining were given.

  • In this study the whole process of pollination an d fertilization in aDole was observed with a fluores.cent microscope, and the thinning mechanism of MCPB—ethyl【4一(4一chl0m一2一methyl phenoxy)butanoate]and CFA (calcium.formic acid compound)as flower thinners Was studied anatomically.It iS proved that MCPB—ethyl and CFA ale entirely diferent in interfering pollination and fertilization of apple. MCPB—ethyl spray did not interfere in the rmination of pollen,the elongation of pollen tube in style and to ovule an d fertilization,but the development of ovule Was restrained to result in fruit drop. On the contrary,CFA did not afect the development of ovule.Fruit drop Was res~ted due to injury of stigma,pollen on stigma,and polen tube in up style,and fertil—ization Was prevented by CFA treatment.

  • AFLP(Amplified Fragment Length Polymorphism)analysis system was established on Ponc/rusaccessions.Seven p1inler pairs th hish polymorphism have been screened from 40 pemer pairs.The screenedsubset of primer paiIs produced 539 bands, 146 of which were polymorphic. Genetic similarity among 36accessions ranged from 0.23 to 0.78. A儿the 36 accessions could be clearly diferentiated based on the bands obtained from three of the seven primer pairs.1his provided a first example to distinguish all tested Ponc/nts
    accessions.The genetic distance was calculated and a dendrogram was constructed based on AFLP data of 7 primerliirs.Th e separation of Poncirus accessions did not correspond to their geographical distribution,which may result from the frequent exchan ge of seeds of Poncirus plan ts among diferent regions recently.

  • Identification and classificati0n of 39 litchi varieties were studied based on A兀P analysis.Threepair of primers(EcoRI AAC+MseI CTG,EcoRI AGC+MseI CAT,EcoRI ACC+MseI CAT)were screenedfrom 64 pare ofprimers.rI1le an alysis ofgenetic diversity among litchi varieties indicated on morphological level lessabundant than molec~ar leve1.The cluster analysis results showed that the 39 cuhivars could be divided into 6groups.It is diferent from the traditional classification system based on peel morphology including peel smoothness,scale size.scale arrangement.rI1le results showed peel morphology Call not be used for litchi varieties classificationas the only criterion.The molecular marker standard map of the main litchi varieties Was established .Two varieties(Litchi chinensis ‘Guiwei’and Litchi chinensis ‘FeizSxJao’) come from diferent arIea were identified.

  • In this paper we studied the post—harvest physiology of antisense LeETR 1 transgenic tomato fruits. Respiratory rate of transgenic fruits was significantly lower than contro1. Ethylene production peak of transgenic fruits appeared 10 d after contro1. Chlorophyll degration an d lycopene biosynthesis Was inhibited in transgenic fruits.After 30 days in normal room,transgenic tomatos eventually developed an orange colour but never turned red.During the course of ripening,cellulase and pely~ acturonase activity of transgenie fruits wel'B signitieantly lower than control and the firmness of transgenic fruits declined more slowly.These datas suggest that LeETR1 Was closely correlated with the ripe ning of tomato fruits.

  • The combining abilities, heritability, correlation, aggression, and path for parthenocarpy were analyzed according to 10 hybrid CI'OSS made between 3 parthenocarpic lines and 2 non—parthenocarpic lines of eggplan t with half mating design.The results showed that the heredity of parthenocarpy was not correspondent with the additive-dominant mode1. Parthenocarpy was ma ybe controlled by a group of recessive genes as well as influenced by epitasis effect.

  • An AFLP and RAPD genetic map with 352 markers was employed in mapping and analysis ql'gcontrolling heat tolerance in Brassica camistris L. ssp.pekinensis,using composite interval mapping method.Heat damage index in seedlings WaS used as phenotypic value to detect quantitative trait loci.Five ql'g weI'e pped in three linkage groups.Three loci including ht-1, lu-3 and lu-5 showed positive additive efect, while ht-2 and _4 showed negative additive efect. Out of the five rL, pbenotypic variation explained by ht-2 WaS
    the biggest one。and then ht-5.The distance from the nearest flanking markers to ql'g ranged from 0.1 cM to 2.4 cM.These markers provided useful information for molecular marker assistant selection of heat tolerance in breeding programmes of Chinese cabbage.

  • Selected ivy(Hedera nepalensis vaur.sinensis)growing in open field and greenhouse separately as the experimental material,the fluidity an d the composition of fatty acids in phospholipids isolated from the leaves
    were determined by fluorescence polarization,scanning thin—layer chroma tography and gas chroma tography methods.The results showed that ivy growing in ope n field in January possessed the higher level of unsaturated phospholipids (PC and PE)and fatty acids(16:1+18:1+18:2+18:3)than that growing in greenhouse. For ivy g∞ in open field,phosphatidyl—choline (PC )Was the main factor to influence its membralle—lipid fluidity during it
    adapted to the low—temperature stress. By an alyzing the diferences of fluidity an d composition of phospholipids from the leaves of ivy growing in diffe~nt temperature conditions, the mechan ism of the cold—resistance of evergreen broad—leaved plant in terms of the composition and the structure of membrane—lipids were discussed.

  • The 2413 was an open pollination hybrid progeny germplasm that derived from branch variance in
    diploid euhivars 75—2—200,which were found from the hybridization between diploid euhivars‘1'ong}nla 3’and
    ‘Shuangqing’. Morphologie and histology characteristics of the 24 13 gennplasm an d cytology identification of the
    root apex were studied in comparison with that of diploid gennplasm 75—2—200.Results showed that 24 13 germplasm
    had larger and thicker leaf, stronger petiole and branch, larger stomata size, smaller stoma ta density, L~,'ger
    pollen size,lower percentage of polen and chrolnosome number Was tetrapolid (2n:4x=76).It is,therefore,
    tetraploid.

  • The variations of style auto—fluorescence after self-and cn)ss—pollination in serotina Rehd.were studied with laser scanning confocal microscope(LSCM).The pollination combinations were as follows:‘Housui’(S3S5) × ‘Housui’ and‘Housui’ × ‘Nijiseiki’ (S2S4). Ihe style auto-fluores—
    cence increased after pollination and the most remarkable increases occurred in the stigma an d the a瑚0—400 pm down(total length of style is about 3700/nn).After self-pollination.with a rapid decline of auto-fluorescence from stigma to 950 tan down,there was almost no variation from 950 tan to the end of style. But a visible climbing of auto-fluorescence can be detected from 950 tan to the end of style after cro~s—po llination. Results also showed
    that there were diferences of style auto—fluorescence distribution between self-and cIDss—po lination treatments.AⅡtreatments proved that in the upper half of the style the auto—fluorescence in the parietal part was stronger than that in the middle part and so did that in the lower part after self-polina tion. But after cn)ss—polination the au to-fluo rescence was relatively evenly distributed in the lower half of the style.All above indicated that,with LSCM ,visible changes in the style after various pollination treatments Can be detected and thus another method to study the self-incompatibility in vivo or senti in vivo Can be established.

  • The results of research shows that the net photosynthetic rate(Pn),stomatal conductance(Gs),intracellular concentration of C02(Ci),Ch1.a and Chi.b content,Chi.a/b and RubisCO content in grape leaves are decreased under high humidity and low light intensity.Both stomatal conductance(Gs),intracellular concentration of CO2(Ci)and RubisCO content are significantly positive correlation with the net photosynthetic rate(Pn)(r=0.9772,r=0.9151,r=0.9101).During the recovery,under slightly high humidity in combination with low light intensity,Pn,Ci,Gs and RubisCO content almost reach their values similar to the contrast level respectively.Using higher humidity in comb ination with lower light intensity,their values could not be recovered.

  • A special inducing device for fruit sunburn was made to study the“threshold temperatures’’with
    attached fruit.The results showed that,the device worked perfectly in calm ,clear and sunny days.Th e diference between~tual average fruit temperatures during induction and designed temperature Was less than 0.12~C in most cases and the fluctuation of temperatures during treatment Was less than ±0.5℃ .]'his technique could meet the
    requirement for field induction of fruit sunbum.Th ere ested some differences among 9 cultivars examined in resistance to thermal injury,ranging from (45.9±0.5)oC to(48.5±0.5)oC.Light Was important in inducing fruit
    sunburn because at the sane“threshold temperature”, fruit got sunburned under natural light but didn’t necessarily without sunlight.

  • The experiment conducted with Red Fuji apple(Ma/us dom/st/ca Borkh)showed that in vitro activities of both a—amylases and amylases of apple fruit significantly increased when adding ATP into the reaction of amylase assay,and this amylase activation was shown to be ATP dose—dependent.W7,an inhibitor of calcium-dependent protein kinases.significantly inhibited this am ylase activation also in a dose—depe ndent.These data indicate that a—amylases and t3-amylases ale activated by protein phosphorylation in developing apple fruit.

  • Sugar contents and activities of sugar-metabolizing enzymes in the peel and juice sacs were analy during the fruit development of Newhall orange(Citrus sinensis Osb.)and Eureka lemon (C.1imon Bunn.f.).The results showed that,1)changes of the sugar contents and activities of sugar-metabolizing engynlL were diferent in citrus cuhivars and in diferent fruit parts;2) the reduction sugar content was higher than the sucrof content in both peel and juice sacs of om ge or lemon,as was diferent from other’s study;3)the continuous decrease of the soluble acid invertase(SAI)activity during sugar accumulation of orange juice sacs,close to zero at the fruit—harvesting time, implicated that the decline of SAI activity was likely related to sugar accumulation;and 4)the phenomena that the activity of the alk~ine inverse(NI)in lemon juice sacs decreased continuous with fruit ripeness and kept the lowest level at the fruit—harvesting time while as the activity of SM reached a higher level,up to 6 times of that of the other enzymes,indicated that the consumption of sugars in lemon juice sacs was related to NI activities during the fruit developmental stages and to SM at fruit maturing period.Sucrose synthase (SS)and invertase might play a role in sucrose metabolism in citrus fruit juice sacs considering the significant diference of sugar contents between in orange and lemon juice sacs as well as the changes of their related crose—
    metabolizing enzymes.

  • The influence of in vitm microenvimnment during rooting stage on the growth and physiology of strawberry(Fragaria al2aglt1.a Duch ‘Fenxiang’)plandets during ex vitm acclinkatization was studied.Nodal cuttings were cultured on solid Murashige—Skoog mediuin with 3%sucrE6e or without sueI'of,and in low (60+10 tanol·m.s )or high(150~10 pmol‘m-2.s )photosynthetic photon flux densities,and normal(350±20 t,-L‘L )or elevated (700±20 t,-L·L )C02 concentration.After 20 days of rooting culture,planflets were transferred to the greenhouse for a month period of acclimatization.Th e results indicate that 3% sucrE6e is be neficial to bimmas forrmtion of planflets,higher PPFD and elevated C02 concentration were both of benefit to the growth of anflets.But higher PPFD reduced the c~omphyH content,and a slight negative efect on photosynthesis could be found in the culture conditions of 3% sui'ose without elevated C02.During the acclima tization,photoinhibition was not obvious,and pPSI1 was incnxIsed.Th e distinctive efects of the diferent in vitro culture conditions on growth.pigments and photosynthetic parameters were decnxIsed during greenhouse acclimatization.Th erefore.we could conclude that pmper sucn)se concentration ma y promote the biomass forrmtion and weaken the sensibility to the envimnnnt variable.and properly elevated C02 and PPFD may be helpful to the growth of plantlets.

  • Four pak—choi cultivars with diferent nitrate accumulation level were applied for explaining their genotypic variation in nitrate accumulation. Their dynamics parameters in nitrate uptaking , Miehaelis constant(Km),the maximum uptake Velocity(V ),nitrate content in nitrate maetabolie pool(MPs)and nitrate re.ductase activity(NRA),were measured,results showed that Km and Vnmx,MPS,NRA varied greatly in diferent oak-choi cuhivars.But there was no relationship between nitrate content an d NRA,MPS. Th eir variation in
    nitrate accumulation more resulted from their diferent uptake rate of nitrate than their NRA,M_lX5.

  • Field surveys and data analysis were used to investigate the primrose rmplasm resources occurring in the Wuliangshan Mountains,South—Central Yunnan.Totally 20 primrose taxa(including species,subspecies and varieties)were recorded in the region.Their distribution patterns and habitats in the
    research regioll were observed and described.The ornamental features (e.g. flowe~,leaves and blooming seasons)of solne important taxa were provided in the present paper.Suggestions to conserve and sustainably
    develop the primrose gemlplasm resources in the Wuliangshan Mountains were also proposed here.

  • T0 investigate possibility of bulblet production of f2ium ‘Casa Blanca’ in bioreactors, bioreac.tor and solid culture methods were studied. Increased bulblet growth was observed in bioreactor culture than solid cultures.Bulblet growth was 16.5 fold in solid culture and it Was 22.4 fold in bioreactor culture. Culture density in bioreactor culture Was studied and 4o0 bulblets were suitable for 5 L bioreactor and from these cultures we obtained 295 bulblets of size≥ 1.1 g. Stem emergence of bulblets obtained from both cultures showed that 90% 0f bulblets(≥1.1 g)from bioreactor culture and 65% from solid cultures(i>2.1 g)developed stem respectively.

  • Preliminary Study on Tissue Culture of Bermudagrass and Its Dwarf Sonmclonal rianth Shaoy, Guo Zhenfei,and Chen Yongchuan(Laboratoryfor Ttwfgrass Science,College of Science,5China,510642,China)The stolonous nodal segments of hybrid bermudagrass cuhivar‘Tifeagle’(Cydon dtylon×C.tranmxta&nsis)were sterilized by 0.1% HgC12 and placed on solid medium contained N6 macronutrient elements,B5 micronutrient elements, MS ferric salt, Gamborg’S vitamin and additives including 2 mg.L 2,4D and 0.02 mg.L 6一BA for induction of callus.The compact embryogenic calli were obtained an d were highly regenerable after they were transferred to regeneration medium.Somaclonal variation was observed among the plantlets.One of them, T、,4, showed dwarfism variation. Compared to its parent ‘Tifeagle’, T、 showed significant morphological variation with decrease of 47% in stolon intemode length, increase of 30% in blade width ,and reduction of 32% in blad e length.TV4 had extensive root system with increase of 37% in dry weight of the newly developed adventitious roots after the stolon was planted for 40 days.TV4 showed higher chilling tolemace than ‘Tifeagle’with lower relative electrolyte leakage after the plants were subjected to 8℃ .SOD activity decreased in ‘Tifeagle’,while that increased in TV4 after chilling treatment.Th e activities of CAT an d POD dechned in both TV4 and ‘Tifeagle’.but they Were maintained at higher level in Tv4 than ‘Tifeagle’.

  • Genomic in situ hybIridization(GISH)was used to discriminate the diferent genomes in the BC1 progeny of Oriental lily and Asiatic lily. We used gen c DNA of Oriental Lily as probe an d HS sperm DNA or genomic DNA of Asiatic Lily as block DNA for chromosome painting in the present paper. 卟e result clearly showed the diference between the two genomes of Oftental Lily an d Asiatic Lily in their BC1.This confirmed that GISH could be used not only to identify the successfulness of the cross between Oriental Lily and Asiatic Lily but also to analyze the chromosome crossover or recombination between the two diferent genomes.

  • Many proteins involve fruit ripening,of which a class of protein called expansin is a new diseovered member.During fruit ripening modification of the primary cell wall is required. Expm~in, without enzyme activity.but has been shown to promote cel wall loosing. There are evidences expansin binding in cellulose mi·crofibrils disrupting the hydrogen bonds formed with xyloglucan during fruit ripening. In order to let more people to know the new aIea, its cutrent status and developing trands were reviewed . Th e discovery, mechanism, gene familv and function of expansin were introduced .Th eir role in the fruit ripening was discussed. Correlation wi the cell wall enzymes of expansin was suggested. New strategies using transgenic technology to regulate fruit ripening were proposed.

  • ‘Yunhong 1’is a very nice selection of red pear from the local res0咀lrces in Yunn~ province.This variety has 0d characteristics such as early bearing,hish product, red skin and 0d l~ldng.The fruit shape is good and the flesh is crisp,juicy and sweet.It has hish q~ ity(SSC 13.5% ,"IS 7.43% , Ⅸ 0.34% ,and vitamin C 31.2 me,/kg)and ,od adaptability to soil,resistance to pear scab and pear eallker,and tolerance to storage.Fmit harvested sea~Dn is from the end of September to the early of October in Kunming.The shelf life 0f fnlit is about 60—70 days.s variety might be grown in the of 1 700—2 200 m altitude in Yunn.
