  • The Kinetics of Calcium Uptake in Apple Rootstock Roots

  • The experiment were carried using apple seedlings(Malus hupenensis Rehder)and pot—cultured three—year old apple tree(Fuji/Malus hupenensis Rehder).It was studied that the kinetics of Ca2+ uptake and the effect of temperature and metabolic inhibitor 2,4-Dinitrophend(2,4-DNP)on Ca2+ uptake rate. The result show that the relation of Ca2+ uptake rate and Ca2+ concentration fits into Michaelis-Menten model under low concentlation of Ca2+ (0.02—0.5 mmol·L-1).The Ca2+ uptake rate rise linearlv with the increase of Ca2+ concentration in both intact roots and excised roots and the rate of Ca2+ uptake in intact roots exceeds in excised roots under high concentration of Ca2+ (0.5-5.0 mmol·L-1).The rates of Ca2+ uptake were inhibited by 2,4-DNP both in intact roots and excised roots.The inhibition rate was bigger under low concentration than high concentration and in excised roots than in intact roots. Under low concentration,the rate of Ca2+ uptake Was influenced markedly by temperature.The α value and Ca2+ uptake rate in excised roots were the highest under 15—20 ℃.Under high concentration,the rate of Ca2+uptake did not afected markedly by temperature.The Ca2+ uptake rate in absorbing roots was similar to extensive roots according to roots surface area.The α value in
    ligneous roots was 54% to absorbing roots and extensive roots.

  • The effect of two pear rotstocks ‘Calleryana’ and ‘Betulaefolia’ on photosynthetic properties of young ‘Hosui’ under salinity was measured by portable photosynthesis systems CIRAS-1.The results showed that:(1)There are obvious discrepancy between Pyrus betulaefolia and P.calleryana efect on the photosynthetic properties of Hosui blade; (2)Pn,Gs,Tr,and CE were higher in Hosui combined with P.betulaefolia (H-B)than in Hosui comb ined with P.calleryana (H-C)under the same conditions,H-C with higher Ci than H-B under high salinity.(3)Photosynthesis of pear blade was inhibited by salinity most cause for stomatal limitation under low salinity at early time,and most cause for nonstomatal limitation under high salinity and low salinity at anaphase;(4)In anaphase oftreatment,the discrepancy between P.betulaefolia and P.calleryana efect on photosynthetic properties was dimi nished under severe salinity.

  • Strawberry and tomato fruits harvested at white and green mature stages respectively were investigated in relation to ethylene production rates and calmodulin (CaM) contents.The fruits were treated for 12 h with exogenous ethylene in combination with verapamil (Vp) or chloropmmaize (CPZ) and trifluoperazine (TFP),and then removed from the ethylene for strawberry.The results showed that exogenous ethylene could increase ethylene production of strawberry by420%-73% and resulted in a peak of ethylene production of toma to fruit with 2 days in advance. Ethylene also increased the CaM contents in both fruits. Vp,CPZ and TFP restrained the ethylene production in strawberry fruit when ethylene applied to the fruit,which indicated that Ca2+-CaM signal
    system maybe involved in ethylene autocatalysis. Vp,but CPZ and TFP, can inhibited the ethylene autocatalysis of tomato fruit,which suggesting that,in contrast to strawberry,ethylene autoeatalysis of the fruit was related to
    Ca2+ influx from the extracellular space, but to CaM .

  • Two LEAFY like cDNA fragments of in length,AFL1 and AFL2 were cloned from the terminal buds of bearing shoot of apple trees.Sequencing analysis showed that the coding regions of AFL1 and AFL2 had 90% homolegy,but the 3’non-coding regions had less than 60% homology.The deduced amino acid sequence of AFL1 or AFL2 cDNA had over 70% similarity with PTFL PEAFLO and ALF genes of poplar、pea and petunia, and less than 70% similarity with TOFLFLO and LFY genes of tomato, antirrhinum majus and arabidopsis respectivehy. RT-PCR analysis showed that AFL1 expressed only in the telminal buds during the transition period from vegetative to reproductive growth; whereas AFL2 expressed in the sepal, ovary, root, stem, leaf and the terminal buds between June to Oct. These results might suggest that AFL1 and AFL2 had a common origin, and their functions had diverged, so that they appeared to play independent roles inⅡ0Ial and vegetative tissue development.Southem blot analysis showed that there were at least two copies of LEAFY homo logues genes in Malus species and pear.

  • The study is mainly on the genetic diversity and relationship of Longan (Dimocarpus longan Lour.) cultivars by applying AFLP (Amplified Fragment Length Polymorphism). 103 polymorphic distinct bands were obtained with two pairs of primers(EcoRⅠACT+Mse ⅠCTT and EcoR ⅠACT+Mse Ⅰ CTC),which were enough to distinguish the 46 cultivars from each other. UPGMA analysis showed that 46 cultivars were divided into 11 groups.The ratio of polymorphic rate varied from 0.2703 to 0.4955.The result showed that the Fujian Dongbi
    was diferent from Guangdong Dongbi and they were different types from Kaiyuansi Dongbi.Zaoshu 1 was supposed to be a new type.The results supplied theoretical basis for cultivar classification.

  • The fruit of Kiwifmit (Actinnida deliciosa ‘Bruno’) was used to investigate the regulation of 1·MCP on fruit ripening and softening at 20 ℃.   • The changes of endogenous polyamine levels induced by high temperatures were investigated in tomato plants with diferent heat-tolerance. Plants were treated at temperatures of 25/20oC (day and night),33/25 ℃ and 38/28℃ at seedling or blooming stage. The results showed that the levels of spe rmidine and spermine in heat-tolerant varieties significantly increased with increase of growing tempe rature. and the maximum was observed at 38/28℃.In contrast, in heat-sensitive varieties,the highest levels of spermine and spermidine occurred at 33/25℃. The temperature treatment of 38/28℃ remarkedly decreased spermine and spermidine contents both in leaves
    and flowers compared with 33/25℃ in heat.sensitive varieties.The putrscine content showed no regular changes both in temperature treatments and varieties. The involvement of polyamines in tomato heat tolerance is discussed.

  • The germination character of wasabi seeds an d efect of diferent temperature, light and plant hormones, chemicals on germination percentage of wasabi seeds were studied. The results showed that germination percentage was the highest under 15 ℃,and germination percentage under 20 ℃ was lower than that under 15℃. 6-BA,GA3,and BR could increase germination percentage of wasabi seeds among all treatments,6-BA acted the most efficiently.When seed coat was removed,germination percentage might arrive 98.0%,and germination speed became quickly,so seed coat could inhibit germination of wasabi seeds.

  • The single node segments of planflets of potato cultivars ‘Mim’ and ‘E-potato-1’ were cultured with salicylhydroxamic acid,the inhibitor of LOX,to identify the effects of LOX on tuberizafion in vitro. The resuits showed that activity of LOX in both the planflets and tubers as well as the concentration of jasmonic acid (JA)
    were decreased markedly in the treatments with inhibitor, accompanying by the declines of percentage of tuber formation by planflets, the number of tubers per plantlet and nlean tuber nlass.The activity of LOX in stolons was the highest. It indicated that LOX plays important roles in tuberization and tuber bulking of potatoes, possiblely through the regulation of JA biosynthesis.

  • In this study, 20 materials of subgenus Yulania (include species and cultivars)were analyzed using random amplified polymorphic DNA markers. 15 primers were screened from 55 arbitrary 10-mer primers, and a total of 274 DNA bands were amplified,262 of which (95.6%)were polymorphism.According to the data, a tree diagram of phylogenetic relationship was constructed using UPGMA in MEGA.The results showed that the gene background of sub genus Yulania is very complex and it was difficult to be classified. So know that the resource of subgenus Yulania in China were abundant.

  • Relative water content and relative electrolyte leakage of three warm-season turfgrasses, Tifdwarf bermudagrass(Cynodon dactylon×C.transvaalensis), manilagrass [Zoysia matrella (L.)Merr.]and carpetgrass [Axonopus compressus (Sw.)Beauv.] were measured after they were subjected to soil drought stress
    The data showed that Tifdwarf and manilagrass had high levels of relative water content (RWC) and low levels of relative electrolyte leakage(REL), whereas carpetgrass showed rapid decline of RWC and rise of REL under soil
    drought stress. The results indicated that the drought tolerance decreased in following order: Tifdwarf > manilagrass>earpetgrass. Proline was accumulated in above three species with a negative correlation to drought stress, indicating that proline accumulation in turfgrasses was resulted from the injury by drought stress. Carpetgrass showed continuous increase in superoxide dismutase (SOD) activity,a transient rise of catalase(CAT)activity and reduction of peroxidase (POD) activity,which may lead to accumulation of H202 and therefore injury to plants. Tifdwarf and manilagrass had a transient rise of SOD activity,and prolonged increase in aetivities of CAT and POD. The results indicated that antioxidant enzymes were associated to drought tolerance of warm-season turfgrasses.

  • It was suggested that litchi fruit growth could be reasonably divided into two stages(Stage I andStage II),and stageⅡ could be further subdivided into two substages(Stage II a and Stage II b).Stage I,which constitutes about two thirds of the whole fruit growth cycle(0-53 DAA),was the phase mainly characterized by the growth of pericarp and seedcoat,during which the influx of solutes was slower than the influx of water into the seed,and it Was almost the same case for the pericarp.Stage II a was the phase mainly characterized by the growth of embryo(53—67 DAA)and the influx of solutes Was faster than water influx into the fruit during this stage.Stage II a was not distinct in fruit with aborted seed.Stage II b(67—88 DAA)was the phase characterized by rapid aril growth and maturation,when the infl ux of solutes Was slower than water infl ux into the aril and the ma turation progressed rapidly.Clustering analysis results of changes in fruit dry mass confi rmed that this revised
    division of two developmental stages Was in accordance with the ma thema tical pattern and wi th the notion of“Ballskin versus bladder efect”proposed by Huang and Xu(1983).

  • Efects of Ectomycorrhizal Fungi on the Growth and Nutrient Uptake of Chestnut(Castanea mollissima BL.)

  • In order to understand the the effects of ectomycorrhizal fungi on the growth and phosphorus nutrition status of chestnut seedlings,experiment using three-compartment mesh boxes were carried out under controlled conditions.Two chestnut seedlings were transplan ted in the central compartment and inoculated with two ectomycorrhizal fungi Boletus edulis or Suillus luteus or not inoculated. The result showed that the contribution of extmradical mycellium to the total uptake of P in plant were 39%-54% in Boletus edulis and 15%-35% in Suillus luteus . Comparing with noninoculated treatment,inoculating with both fungi decreased Olsen-P and pH of soil in hyphae compartment.which indicates that both fungi aIe able to mobilize phosphates that are seareely-soluble and low availability in the soil.It is concluded that both ectomycorrhizal fungi play crucial role for P nutrition and the growth of chestnut.

  • Photosynthetic characteristics were carried out with Rubus strigosus Mick.The results indicated:the curve of diumal variation of Rubus strigosus Mick. leaf photosynthesis showed two peaks,appeared ‘Wuxiu’ phenomenon.Pn was positively significantly correlated with Gs,at the same time,the variation of Pn,Gs and Ci was synchronism; the principal environmental factors,which efected Pn,were PFD and RH;the curve of seasonal variation had two peaks; The light compensation point of photosynthesis and light saturation point of Rubus strigosus Mick.1eaves were 750 μmol·m-2·S-1and 21.4 μmol·m-2·S-1, respectively.

  • Using the shoot—tips as the explant,the tube propagation of Prunus pseudocerasus was studied. The optimal asepsis and culture condition were established by a statistical method. The explant was asepticized as follows: 70% ethanol 30 seconds→ 0.1% benzalkonium bromide solution 15 minutes→0
    .1% HgCl2 3 minutes, besides 50 mg/L carbenicillin must be added to the media.The optimal culture condition as follows:(1)the medium for primary propagation contained F14+6-BA0.5 mg/L+IAA 1.0 mg/L+GA30.5 mg/L,; (2)the medium for multiple propagation contained F14+6-BA0.5 mg/L+IAA 1.0 mg/L+GA30.5 mg/L, 2%sugar and 0.5% sorbitol.The results also showed that diferent explants. the length of shoot-tip and the application of anti-oxidant could afect the surviving rate.

  • After 10 h illumination under the light of 50 μmol·m-2 ·s-1,obvious speckles could be formed in the place where the Methyl Viologen was dripped as little blobs on the surface of banana leaves. By determining the activity of two chief antioxidative enzymes,superoxide dismutase(SOD)and catalase(CAT),a significant reverse linear relationship was found between the diameter of Methyl Viologen speckle and the two enzymes activities, showing the close relationship between the speckle diam eter and the antioxidative capacity in bananas. Exogenous active oxygen species could induce the damage such as peroxidafion of membrane lipid in banana leaves , the damaging degree is closely related with the diam eter of Methyl Viologen speckles,this result indicated that the rapid method to evaluate an tioxidation capacity in bananas was feasible.

  • Callus of anther from strawberry (Fragaria ananassa Duch. Darslect) was induced on MS medium. Late embryogenesis abundant protein gene,LEA3,from barley(Hordeum vulgare L.)was introduced into strawberry callus by the aid of biolistic-mediated transformation method.After 4 times of PPI’selected culture. resistant calli and regenerated plants were obtained.The hybridization bands were determined in diferent transgenic plants by Southem blot,and this showed that LEA3 gene was integrated into strawberry genome.

  • A study of inducing polyploidy by treating ‘Luotian Tianshi’persimmon in vitro leaflet explants with colchicine solution was perfonned.The results indieated that mutated planflets can be induced by treating the leaflet in 0.3% colchicines solution for 4—6 d.Stomata of the mutated planflets were significantly larger than those of the normal planflets.but the density of stomata were reduced signmcantly.The chromosome number of root—tip cell of mutated plantlets was 2n=12x=180, and nuclear DNA content of the mutated planflets were twice as much as normal hexaploid plantlets.These results suggest that the ploidy of mutated planflets was autododecaploid.

  • Morphological characters of the pollen grain of diploid, triploid and tetraploid watermelons (Citrullus lanatus)were observed with the aid of scanning electron microscope.Population polen of the diploid waterm elon were in good order; their pollen’s shape with 3 apertures was subprolate and average P/E ratio was 1.283;The polen grains of the tetraploid was spheroidal with 3,4 and more ape rtures, average pollen’s P/E ratios was 1.111; Heteromophic, adhesive and shrivelled pollen were observed on tetraploid watermelon.All of the pollen grains of the triploid were shrivelled . The reasons of lower germination of antotetraploid pollen and abotive pollen of triploid were discussed from palynological observations.

  • Four strains used as female parents were crossed with 8 selflines separately, 26 triple hybrids were obtained. The variation degree of main characters, diferent significan ce of triple hybrids, the relevance of fruit shape betwen the triple hybrids and its parents’ were all analysed.The results show:the variation degree of fruit mass of triple hybrids Was the biggest(variation coefficient was 15-28);then was the sugar (5-16);the fruit shape was relatively stable (<10),the diference of its variation degree and the double hybrids was not
    significant,the fruit shape of the triple hybrids was relevan t with its male parent; the variation degre of blossom stage in the same triple hybrid Was very small.

Makino. var. communis] tended to decline, the weight-loss increased rapidly,and respiration rate showed a climacteric-like pattern with climacteric on 6th day in the duration of storage under the conditions of (20±1) ℃。85%-95% relative humidity.The content of chlorophyll,soluble proteins and carotenoid in outer lcaves decreased more rapidly than that in inner lcaves during storage.In outer leaves there were marked chan ges in
    supemxide dismutase(SOD)and catalase(CAT)activities and the activities of the two enzymes decreased rapidly after 4 days of storage with a mole obvious decrease in CAT activity.Pemxidase(PoD)activity and malondialdehyde(MDA)content increased gradually and then rose sharply after 6 days and 4 days of storage, respectively,indicating an inbalanced metabolism of activated oxygen occurred and resultant accumulation in activated oxygen was
    associated with membrane lipid pemxidation.In inner lcaves the activities of both SOD and CAT were at high level and varied very little while POD activity didn’t increased significantly。but MDA concentration declined a little. These data indicated a balance between the production and elimination of activated oxygen existed in iBrier lcaves prevented the accumulation in activated oxygen and the progress of me mb rane lipid pemxidation. It could be concluded that the senescence of B.chinensis var.commumis was caused by maladjusted metabolism of activated oxygen with outer leaves being senesced much easier than inner leaves, therefore, the key factor for storage of Chinese cabbage is to delay the senescence in outer leaves.

  • Molecular test and insect bioassays with aphid (Myzus persicae Sulzer) larvae verified that gna gene had been integrated into Chinese cabbage genome mediated by Agrobacterium tumefaciem ,and all 7 transgenic plants were aphid resistant,strong mRNA of gna could be detected in transgenic plants with higher insect resistance. The results of inheritance analysis showed foreign gene in 2 transgenic Chinese cabbage plants 282-3 and 282-9 is in the model of the simple Mendel’s fashion in their T1 progenies.

  • Induction, diferentiation and rooting of different explants of Pteridium apuilimun were described on different medium in which contained different hormones and a high egicient plant regeneration system was established. The result showed that the most suitable medium was 1/2 MS+0.2 mg/L+IBA 0.4 mg/L+NH4H2PO4; the induced frequency of bud from callus was 100% on medium in contained 1/2 MS; The radiation rate of adventitious seedling was 100% on medium 1/2MS + 0.2 mg/L IAA, simultaneously vigorous rhizome was formed and the survival rate of transplantation of test-tube seedling could reach a high level at 94%.

  • Flower bud of Daphne odora var. narginata Mak. turns from terminal bud. Its diferentiation includes two processes of diferentiation of inflorescence and of floret, can be divided into seven developmental stages:undiferentiation, different beginning, differentiation of inflorescence, floret, petal, stamen and pistil.   • Flowers with branches of cut rose ‘Belami’ (Rosa hybrida ‘Belami’),a cultivar with middle range tolerance to water deficit stress, were treated with 30 h water deficit stress at room temperature and 40%relative humidity. Then the flowers were reput in water, and vased continuously until loss of vasing value.
    Changes of the contents of soluble protein and free amino acid, and endopeptidase activity in petals were determined. The contents of soluble protein and free amino acid were increased obviously in treated petals as
    como.-,~1 with untreated contro1. and the efect Was lasted even after rehydration and during flowers vasing.
    Endopeptidase activity of petals in treated flowers Was much higher than in control immediately after treatment,
    however the activity in treated flowers Was lower than in control at pH6 determination condition although it Was
    higher at pHlO after rehydration.A new kind of endopeptidase might be induced by the treatment.

  • Mature caxyopses of two materials of Zoysia japonica Stend.coilected in diferent habitats were used as initial explants for callus induction. MS containing 2.0 mg/L 2,4-D induced calli at the relatively higher percentage of 31.2%-42.1%. Inclusion of 6-BA in callus induction med ium with various levels of 2,4-D made an important role for non-embryogenic calli changing into embryogenic calli.The embryogenic calli of higher percentage were induced by a combination of 2.0 mg/L 2,4-D and 0.1 mg/L 6-BA.The rates of shoot regeneration and rooting were different in different regeneration medium. The rates of shot regeneration and rooting were 46.8%-48.1% at the level of 0.1 mg/L 2,4-D.

  • A study on tissue culture of gladiolus hybridus of ‘Chenghongjiao’ and ‘Ziyinghua’ was carried out in MS medium with diferent phyto-hormone BA,KT and NAA.A MS medium supplemented with BA 0.5 mg/L+NAA 0.5 mg/L+KT 1 mg/L was for the regenerated and 1/2MS medium without hormone was for bulbs forming.The three main viruses of TMV,CMV and TVY in gladiolus were removed from cultural plant of gladiolus in the regenerated medium through 5-20 mg/L concentration of ribavirin,treatment of high temperature 38-40℃
    and 2 times of meristem-tip culture.

  • 1-MCP is an effective ethylene action inhibitor and could delay the senescence, improve the storage quaity, promote the shelf life and affect both the flavors and physiology disorders in postharvest fruits. 1-MCP also could regulate some related some related enzyme activity and related gene’s expression in fruit senescence. This paper reviewed these previous studies on the effects and physiological mechanism of 1-MCP in delaying fruit senescence, In addition, the potential with an emphasis on applications of the research in this area are proposed.

  • ‘Jiaxian Youzao’,a new drying variety with late repening,selected from ‘Zhongyang Muzao’ jujube orchards in Jiaxian county,Shaanxi. The fruit is oval or column shape,red-black colour, and middle large, the average fruit mass is 12.3 g,maximum fruit mass is 19.6 g.It ripens in early-middle October in north Shaanxi.The fruit is uniform, attactive, good quality fruit, and applicable to dry. Its cultivated region is in north China with the cool temperature (9-11 ℃) and less rainfall (annual rainfall of 390-500 mm).

  • ‘Zhangiiao 8’ is a fine promoted variety,which is breeded and selected from mutated plants in vitro tube-buds of Fujian ‘Taiwan Grand Cavendish’ irradiated by 60Co γ ray.The variety grows up with characters of its pseudostem light green,petiole longer and plant type straighter,morever without pappus on its fruit axes.Compared with control, the variety has an over 10% increase of yield, finer appearance of fruit ears, longer and heavier fruit fingers.For richer in vitamin C and higher of TSS/TA,the variety tastes more flavorous.
