  • Using seedlings of Malus baccata, M.xiaojinensis and M.zumi as the experimental materials. Na2+ exclusion capability was determined to affirm the whole Na2+ exclusion capability, and the whole Na2+ exclusion capability of salt-sensitive species was about 65%, middle salt-tolerant species about 8O%, salt-tolerant species about 90%. Na2+ exclusion capability of roots and stem base was the main parts of the whole Na2+ exclusion capability in Malus seedlings. Na2+ exclusion capab ility in roots and stem base of salt-tolerant species was obviously higher than that of salt-sensitive species. Lateral roots.stem base xylem and old leaves of salt-tolerant cuhivar obviously occured Na2+ accumulative effect, and were main Na2+ accumulative localizations. Through shoot Na2+ re-distribution and re-transportation. Na2+ content became even less in young leaves an d mature leaves of salt-tolerant species

  • Changes of endogenous ABA, GA3 content, ABA/GA3 value in sweet cherry flower buds and effects of correlative environmental factors were investigated during dormancy on 8-year-old ‘Jueze’ (]h4IeMMa)and ‘Zao Hongbaoshi’(Py6Hnoaaa) sweet cherry trees with HPLC (High Performance Liquid Chromatography).It was found that ABA content rose steadily in primary stage and dropped in later stage during dormancy, and its maximum appeared at the begin of November.Contrary to ABA, GA3 content dropped sharply in primary stage of dormancy and rose steadily during release of dormancy, its minimum appeared at the end of November. Moreover, ABA/GA3 value rose with the deepening of dormancy and dropped with the release of dormancy. Temperature affected ABA and GA3 content in dormant flower buds: low temperature(5℃)raised GA3 content and reduced ABA content obviously, moreover, the time when ABA began to fall and GA3 content began to rise advanced after low temperature treatment, which made flower buds release dormancy earlier than contro1. The efect of high temperature(20℃)was contrary to that of low temperature(5℃).Variable temperature(5/20℃)affected dormancy release slightly, but its effect was less remarkable than 5℃ treatment because of the counteract of 20℃. Overshadow treatment before defoliation could reduce GA3 content slightly, but its effect on ABA content was little. The synthesis of GA3 was promoted and synthesis of ABA was inhibited after long photoperiod treatment,contrary to that,short photoperiod treatment raised ABA content and reduced GA3 content sharply.

  • The detailed ontogeny of postgenital fusion within the gycoecuim systematically investigated with light microscopy in ‘Kyoho’ grapevine.The result shows that the two carpels are free at the early stage of floral ontogeny.During normal floral development the two carpels primordial initiated.The separate carpels primordial lateral growth continuously upward with opposing distal tips of the two carpels primary meristem approached
    to each other.The adaxial surfaces of two carpels primordial contacted at upon ovary regions,and the epiderm al cells in the contact zone rapid redifferentiation into parenchymatous cells.As a consequence of this epidermal cells redifferentiation the carpel become fused by the infolding,and this region then proceeds to develop into the style,the stigma,and the distal region of the ovaries.After the completely fusion carpel of ovary formed two locules separated by a septum.Ferthermore,this paper indicated that the ontogeny of three carpels of ovary with two patterns in grapevine.

  • TotM RNA was extracted from aborted—embryo of litchi ‘Guiwei’. Dephosphorylating RNA,removing the mRNA cap structure and ligating the RNA Oligo to decapped mRNA with GeneRacerTM Kit and then the first strand of cDNA was synthesized by reverse transcription. The 3’and 5’gene specific primers from differential expressing cDNA fragments( gene),isolated from aborted-embryo of litchi varieties of Guiwei by SSH,was designed with Gene Tools.The full-length 3’and 5’ends of cDNA was amplified with the first cDNA as template and two gene specific primers or nested primers by PCR. Complete SAM gene sequence was obtained by BLAST comparison of the three fragments and splicing according to the overlapping regions. The sequence of the SAM synthetase gene is 1515 bp in length with an opening reading frame(ORF) encoding 393 amino acids.The nucleotide sequence exhibits about 79%—82% overall similarity to the corresponding gene of other organisms,and the amino acids is about 88%—91%. It was suggested that
    the obtained cDNA was a SAM gene in aborted-embryo of litchi varieties of Guiwei.

  • Change of Mineral Nutrient Elements Content in Growing Period of Cerasushumilis

  • The changes of content of several mineral nutrient elements,including Cu,Zn,Fe,Mn,Ca,Mg,K in leaves,flowem and fruits of Cerasus humilis growing period were analyzed’vith an atomic absorption spectrophotometer and the correlation among the changes of these elements was also studied.The reset showed that the flowering phase of Cerasus humilis was the key period for the absorption and cumulation of mineral nutrient elements. In the whole phase from start to end,the increase of content of element Fe,Ca, Mn Was remarkable, but there Was no evident increase or decrease for that of other elements.After flowering phase,the content of seven mineral elements keep hish in former period and low after that. There appeared a changing trend of descend-plane(ascend)-descend in content of all seven mineral element.There was a significant positive linear relationship of content of 6 elements in flower and fruit of Cerasus humilis,except element K. When compared 山that in leaves,only Ca and Mg had a significant positive linear relationship,Zn had a significant negative linear relationship and there were no sign ificant corelation in that of other elements. The sequence of 7 elements by the average content in fruit was K>Ca>Mg>Fe>Zn>Mn>Cn.

  • The Effects of Postharvest Low Oxygen Treatment on the Storage Quality ofChestnut

  • In this paper,the efects of postharvest low oxygen (0—5% )treatment on the cold storage quality of chestnut(Castanea mollissima Blume) ‘Dahongpao’at 0℃ were studied.The results were as follow: (1)Treatment duration and oxygen concentration were the main factors afecting the quality,the chestnut become off-flavor if it was treated for more than 20 days and the oxygen concentrations were below 2%.Though the index of off-flavor was alleviated during the 120 days cold storage,the flavor was not eompletely recovered at the final.(2)Low oxygen treatments induced the increase of ADH activity and alcohol concentration,the longer treatment time and lower oxygen concentration,the higher ADH activity and alcohol concentration.When the treated chestnuts were stored at 0℃ for 120 days,the ADH activity and alcohol concentration were recovered to the level of control.(3)Low oxygen treatments afected the metabolism of starch and soluble sugar.0—2% oxygen could stimulate the decomposition of starch.while 3%—5% oxygen could slow down the decomposition rate. The treatment of 1%—3% oxygen maintained higher content of soluble sugar than the other treatment.(4)The mean decay rate of 0—2% oxygen treatments was 10%,the control was 7%.Meanwhile the decay rate of 3% —4% oxygen treated for 20 days or 5% oxygen treated for 25 days were only 1% at the end of 120 days cold storage.In general,3 oxygen treated for 20 days was the best treatment to keep the storage qu ality of chestnut among the low oxygen treatments.

  • With the increase of soil organic matter,the mineralizable nitrogen,which is about 95 percent of soil total nitrogen,Was also increased,thus soil can provide more nitrogen.In the treatments with application of nitrogen alone or combination with phosphorous an d potassium,it Was dificult for the N sink to be enlarged,SO the available nitrogen Was decreased quickly.Long-term fertilization trial in protected field showed that the soil was rich in total phosphorus an d lack of potassium.Excessive and long-term application of inorganic nitrogen alone accelerated the progress of soil acidization, decreased the exchangeable bases and their proportion was in disorder.Such problems can be prevented by application of organic matter,protected cultivation and comb ination tll phosphorus an d potassium.Soil physical degradation was a long-term progress from quantity change to quality change and it Was destructive to soil productivity.

  • The innuence of cinnamic acid at diferent concentrations(O,80,150 µmol·L )on leaf gas exchange and cell uhrastructure of cucumber seedlings was investigated in substrate culture. Resulted showed that the einnamie acid at 80 µmol·L significantly reduced the leaf-area,the content of chlorophyll
    a,the photosynthesis rate,the respiratory rate,the activity of root and the opening number of stomata,aceompanied by a deformation of the ultrastructures of chloroplast,mitoehondfion.More significa t effects were
    observed at a 150 µmol·L cinnamie acid treatment.All these showed that cinnamic acid significantly influenced the photosynthesis by reducing the activity of root,function of the chloroplast and mitoehondrion,stomata
    operation.

  • With a F2 population between a highly resistant parent and a highly susceptible parent(Q9×Q10)as materials,polymorphism between resistant and susceptible bulk of cucunlber powdery mildew were studied using BSA method and AFIJP technology. A Co-dominant AFLP marker,P18M47—238 bp/236 bp,was screened.The resistant and susceptible F!plants possessed the 238 bp and the 236 bp fragment,respectively, and the mid-plants owned the two fragments.Linkage analysis indicated that the AFLP marker was tightly linked to the cucunlber powdery mildew resistance related gene, and the genetic distance was 5.56cM. Sequencing of the fragments of the Co-dominant AFLP marker indicated that their lengths were 238 bp and 236 bp respectively.The difference of the two fragments lied in the insert-deletion (indel)of two“T”base.BLAST analysis revealed that the two fragments were new in the GeneBank.

  • Cotyledons with petiole from aseptic seedlings of Chinese cabbage (B. campestris ssp.pekinensis,cultivar‘Beijing 80’)were used as explants for in vitro transformation using Agrobacterium tumefaciens C58 carring plamid pBBBasta-pinⅡ-bar.Via series concentration selection of PPr.the transgenic
    resistant plants were got,and showed high Basta resistance when transplanted to the greenhouse.It was proved that by shoot regeneration from young leaf disc of the survived regenerated plant,the chimera status
    of the plants can be eliminated.Transgenic plants were confirm ed by PCR,PCR-Southern,and genomic Southern blotting which showed that the bar and pin Ⅱ genes were co-integrated into the plant genome.Continuous insect feeding trials using larvae of Pieris rapae L.revealed that larvae fed with transgenic leaf tissue had a higher mortality,and the developments of them were retarded compared with those larvae fed with nontransgenic control plants.

  • Four genotypes of pepper were used to study the effects of AgNO3,Vitamin C,carbon source,nitrogen source,harvest time,plant growth regulator on the anther culture of Capsicum annuum L. through orthogonal tests.As the results,AgNO3,Vitamin C can reduce the browning of the tissues;Plant growth regulator,active carbon,AgNO3,Vitamin C have a higher effect on calli yields;Vitamin C,harvest time,carborn source,AgNO3 have a higher effect on embryogenesis.A new modified culture medium was concluded.

  • The effect of 1-methylcyclopropene (1-MCP) on the senescence of broccoli was studied.The results showed that the shelf life of broccoli was extended and the degradations of chlorophyl 1-carotenoid and vitamin C delayed by exposure to 1-MCP at concentration range of 0—20 µL/L for 6 h at room temperature (20 ℃)followed by storage at 20 ℃ 1-MCP at optimal concentration of 2.5 µL/L was used to investigate the possible mechanism of 1-MCP in delaying the senescence. The respiration rate and ethylene production of florets were inhibited, the protein content of florets, the activities of SOD and CAT increased, while the MDA content and the activity of POD decreased during storage by exposure to 2.5 µL/L 1-MCP.SDS-PAGE analysis further showed that the composition and content of both the alcohol-soluble and water-soluble proteins changed after 1-MCP treatment.

  • The experiment concerned the 3 years.old plants of pot azalea (Rhododendron simsii ‘Mission Bell’)were designed as 30 replicates to conduct studies on media formula with hydroponics.In terms of plant height and width,leaf length and width,fresh and dry mass,root development and leaf chlorophyll content(SCDSV),the results indicated that M 1(Peat moss 3+Vermiculite 3+Perlite 3 by volume)formula was the best media for soilless culture of pot azalea plants.the medium formula containing coconut was
    not suitable for soilless azalea culture. By analyzing physical and chemical properties of different media formu1a before and after subirrigation with Sonneveld nutrient solution.it was found that pH values tended to 5 and
    EC values increased a lots as a result of salt accumulation in media at a closed system of hydroponics:all the contents of total N, available P,K,Ca, Mg,and CEC in 5 media form ula increased as wel1. Finally,contents of macronutrients and concentrations of micronutrient in plants of ‘Mission Bell’were almost around the optimal levels of plant growth.

  • The shoots of pear and apple in vitro were irradiated by gamma rays. The effects of gamma irradiation on shoot regeneration from leaf and bud growth were studied. The organ which is the most sensitive to γ-rays was leaf,second was unbudding lateral bud, the third was growing shoot tip. The LD50(50% decrease of regeneration)of leaf was genotype-dependent and was between 20 and 30 grays.The LD50 of lateral bud also was genotype-dependent and was 30 grays or above 30 grays. Doses of iradiation were between 10 and 30 grays,the elongation growth of shoot tip was inhibited, but no lethality or other symptoms.

  • An efficient and simple method for shoots regeneration from in vitro leaves of ‘Gisela 5’ and ‘Gisela 6’ (Pruns cerasus×P.canescens)was developed.Factors influencing in vitro leaf regeneration were studied.The ability to regenerate shoot from in vitro leaves of‘Gisela 6’ was much greater than that of ‘Gisela 5’:Optimal results were obtained by culturing leaves adaxial side down on WPM medium containing BA 5 mg/L+IBA 0.5 mg/L and BA 7 mg/L+IBA 0.3 mg/L respectively in the dark for 15 days.Shoot regeneration rate of ‘Gisela 6’ reached up to 10o% and that of ‘Gisela 5’ was 75.1% .The regeneration shoot was easy to rooting and to transplant to the field.

  • Identification.classification and genetic relationship of 30 litchi germplasms of Hainan island were measured at the molecular level by random amplified polymorphic DNA (RAPD)with ten arbitrary primers of 10 bases. The results revealed that similarity coefficients of most of litchi germplasms were among
    55%—8O%.Which indicated that the genetic relationships between them were closer related. 30 litchi germplasms were divided into 4 groups at the level of the similarity coemcient of 61% .Although a certain correlation with the morphological classification based mainly on the characteristic of tubercles on the ripe fruit skin,the range of classification by RAPD was more detailed than that of morphological classification.It is concluded that the traditional morphological classification system can not reflect fully the genetic relationships between litchi germplasms,and need further improvement according to the result by RAPD analysis.

  • The strawberry fruits(Fragaria ananassa Duch.‘Chunxing’) harvested at white stage were used to investigate the efects of exogenous ethylene (50 µL·L-1 ) on Ca2+-ATPase activities,O production rate and MDA content of microsomal membrane with or without pretreatment of 100 µmol·L-1 chloropromaize(CPZ),trifluoperizine(TFP)and verapamil(VER),respectively,The results showed that the exogenous ethylene had no obvious influence on O production rate,and had no much effect on MDA content,but stimulated Ca2+-ATPase activitv of microsomal membrane at 12 h after ethylene treatment.Whereas a decreasing of Ca2+-ATPase activities, and relatively higher MDA content were found at 24 h after ethylene removal.The pretreatment of CPZ,TFP and VER reduced the Ca2+-ATPase activities and MDA content of microsomal memb rane under ethylene treatment, but had no significant efect on O production rate.These suggested that cellular Ca2+-CaM may be involved in regulation of Ca2+-ATPase activity and lipid peroxidation of microsomal membrane induced by ethylene.

  • Sugar and aroma components in fruits of strawberry(Fragaria×ananassa Duch.CV.Hada) were analyzed under priming leaf treatment.The results were as follow:43,33 and 37 types aroma components were identified in not priming leaf,1/3 priming leaf and 2/3 priming leaf treatments respectively.Aldehyde aroma concentration increased with the increasing of priming leaf severity,while the concentration of ester aroma,2,5-dimethyl-4-methoxy-3 (2H)-Furanone,fructose and total sugars in fruits significantly decreased.

  • Somatic fusion hybrid DSa(S.torvum Sw×S.melongena L.‘Dourga’)microspore were cultured by using isolated microspore culture. The result show that microspore dediferentiation ability was markedly improved by using the treatment of anther under the high temperature conditions of 36 ℃ for
    continuous 6 days. The best development period for microspore culture is monokaryotie stage;The suitable medium for microspore culture is KM with PEG 250 mg/L,2,4-D 0.2 mg/L,ZT 0.5 mg/L,NAA 1 mg/L and glucose 6.5%;The suitable medium for regenerated plantlet is MS with ZT 2 mg/L,IAA 0.1 mg/L,sugar 2% and agar 7 g/L.

  • The effects of root-zone hypoxia treatment on nitrogen metabolism in muskmelon seedlings were studied in a hydroponic culture system.The results showed that roo-zone hypoxia treatment NO3 --N, NH4+-N contents and NR activities in the roots and leaves all increased,the soluble protein contents decreased,the heat-stable protein contents in the roots increased.The increasing degrees in the roots were more than those of leaves.Under the hypoxia treatment,the decreasing degrees of NO3 --N, NH4+-N,soluble protein contents and NR activities of‘Gangqiang’were more than those of‘Xiyu 1’.It indicated that the higher contents of NO3 --N, NH4+-N,protein and NR activity played an important role in enhancing hypoxia resistance in muskmelon seedlings.

  • This paper mainly dealt with the study on shoot organogensis culture in vitro and plant regeneration from sprout explants of Elaeocarpus sylvestris (Lour.)Poir.The results showed that the best media for various stages were as follows:(1) Initial medium:MS+BA 1.0~2.0 mg·L-1 +NAA 0.05 mg·L-1 +3% sucrose;(2)Clump shoot regeneration medium from callus proliferation:MS+BA 1.0mg·L-1 +NAA 0.5mg·L-1 +3%sucrose;Clump shoot regeneration medium from axillary buds:MS+BA 2.0 mg·L-1 +IBA 0.1 mg·L-1 +3%sucrose;(3)Efficiency seedling induction medium:MS+BA 0.5
    mg·L-1 +IBA 0.1 mg·L-1 +GA 1.0mg·L-1 +3%sucrose;(4) Rooting medium:1/2 MS+IBA 3.0 mg·L-1 +2%sucrose. In root induction culture.the rate of root formation with membrane filter is higher than that of root form ation with common membrane.

  • The study aimed on the selection of embryonic callus of Kalanchoe blossbeldiana,the induction of embryoid,morphogenesis of embryoid and regeneration.The grain-like,light yellow and loose callus was embryonic callus. During the inducing of embryoid,the culture mediums with different growth regulator concentration were used.Finally the culture medium with the highest of reduction frequency was MS+BA 2 mg·L-1+NAA 0.2 mg·L-1+Sucrose 30 g·L-1 +Active carbon 4 g·L-1 (185 embryoid per gram).The regeneration medium Was MS+Sucrose 30 g·L-1.The appearance and growing of embryoid were also observed through parafin slices of callus.The development procedure of embryoid Was procel of embryoid,globular stage,heart-shape stage and cotyledonary stage

  • The main factors of crowd shoot induction,propagation and rooting of Oncidium were studied by using different growth period of bud.The results showed that the primary bud is better explants than others.The different kinds and different concentration of hormones greatly affected crowd shoot induction,propagation and rooting of Oncidium.The suitable culture medium for crowd shoot induction is MS+6-BA 4.0 mg/L+NAA 0.5 mg/L +3%sucrose. The best culture medium for shoot propagation is MS+ 6-BA 2.0~4.0 mg/L+NAA 0.5 mg/L+3% sucrose.The proper culture medium for rooting is 1/2 MS+IBA 0.5 mg/L +2% sucrose.

  • ‘Yuxin 1’ iS a new Chinese cabbage hybrid.whose parents are self-incompatible double haploid lines induced by isolated microspore culture. It takes about 75 days from sowing to maturity,its head shape index is 1.28.This variety is multi-disease resistant against turnip mosaic virus(TuMV),downy mildew and black spot.Th e marketable yield is 81.1—90.7 t/hm .It also has excellent quality and good storage tolerance.
