  • Comparison on Starch Characters in Endosperm of Ginkgo biloba var. epiphylla Mak. and Its Systematic Significance

  • The endosperm of normal seed and epiphyllous seeds were determined by Polarimetry and Kjeldahl nitrogen method. The result showed that the starch contents were 63.22% and 61.91%,and the protein contents were 11.35% and 13.36%,respectively. Observation by scanning electron microscopy
    (SEM)showed that most starch grain were spherical and elliptical,which can be divided into three types:A(large grain),B(medium grain),C(small grain). Type A and type B were contained in both normal seed and epiphyllous seeds,while type C was only observed in epiphyllous seeds. The size and the ratio of long axis to short axis of A starch grain in nomal seeds were 14.28 μm × 11.96 μm and 1.24,which in epiphyllous seeds were 13.63 μm × 10.55 μm and 1.30,respectively. The size of B starch grain in normal seed and epiphyllous seeds were 8.29 μm × 7.28 μm and 8.90 μm × 7.68 μm,respectively. The size of C starch grain was 2.07 μm × 1.84 μm. Compared with normal seed,the starch grain in epiphyllous seeds has following properties: different in size, type B and type C starch grain were obvious, the starch grains producted by “gemmation” from large grain, interspaces among starch grain were large, stromatin were abundant, and there were concave field in starch grain. There were significant differnces in starch grain
    types, size, shape and content as well as the stromatin among different individual, especially in the connection mode of starch grain and proteins in normal seeds and epiphyllous seeds and in the stromatin distribution on the starch grain. The low content of starch and various shape of epiphyllous seeds was related to delay development in endosperm and small seed size. The systematic significance of the starch grain in Ginkgo biloba var. epiphylla Mak. was discussed in this paper.

  • Molecular Evolution and Phylogeny of Stone Fruit Trees Based onSequences of the Internal Transcribed Spacers(ITS)of Nuclear RibosomalDNA

  • To analyze the molecular evolution and phylogeny of stone fruit trees,the internal transcribed spacers(ITS)of nuclear ribosomal DNA isolated from 16 genotypes of peach(Prunus persica),plum(P. salicina),apricot(P. armeniaca),mei(Prunus mume),and cherry(Prunus avium),2–4 genotypes per species,were sequenced. All these ITS sequences together with other 6 ones downloaded from the GenBank were analyzed using the second way of rooting with cherry used as the root. The broad data set was tested with 56 evolution models by PAUP 4.08b software,and the scores were used to screen the best model and parameters with Modeltest 3.7 software. Mega 3.1 software was used to calculate genetic distance and variance among the genotypes and to construct maximum parsimony phylogenetic tree. The result indicated: Molecular evolution rates of ITS1 and ITS2 among the stone fruit trees were different,same did the information of them; the evolution order of 5 stone fruit trees was that cherry,plum,and apricot originated from same one ancestry plant,and then plum evolved to peach and apricot to mei;Prunus mandshurica is more original than P. sibirica and P. armeniaca;the evolution order in Prunus persica is from P. andersonii to P. davidiana and then to P. persicaP. ferganensis. It is reasonable that stone fruit trees are divided into 4 subgenera.

  • Treatments of 1 000 nL · L-1 1-MCP fumigation(1-MCP),4% CaCl2 dip(Ca)and the combination of 1-MCP and Ca(1-MCP + Ca)were applied to harvested Huangguan pear fruits. The results showed that compared with control(without 1-MCP or Ca treatment),treatments of Ca,1-MCP and
    1-MCP + Ca maintained the higher firmness,titratable acid(TA)and chlorophyll contents,reduced the skin browning spot index,had slight effect on contents of total soluble solid(TSS)and vitamin C of fruits,also decreased peak value of ethylene production rate more or less,reduced soluble polyphenol content and polyphenoloxidase(PPO)activity markedly, and suppressed the rise of cell membrane permeability and H2O2 content of peel. But it had no obvious effect on the respiratory rate of fruits. Treatments of Ca
    and 1-MCP + Ca significantly increased Ca content of peel and flesh of fruits. On the whole,1-MCP + Ca was the most effective way to keep fruit quality in all treatments. It indicated that inhibiting the production of the polyphenol substances and keeping integrity of cell membrane structure should be an important cause for 1-MCP and Ca treatments, mainly Ca,to reduce the browning spot of peel in Huangguan pear fruits significantly.

  • Eighty plants from the F1 population of Ichang Papeda 2586 (resistant genotype) × Lingnan Shatian Pummelo (susceptible genotype) were inoculated with citrus canker [Xanthomonas axonopodis pv.citri (Xac)] for resistance evaluation in field. The results revealed that resistance segregation in the F1
    progenies was significant, and the ratio of resistant and susceptible plants was close to 1︰1. If immune and highly resistant types were regarded as disease resistance and moderate resistance, moderate susceptible and susceptible were regarded as susceptible types. A SSR molecular marker CCR-110
    putatively related with resistance of citrus canker was obtained based on BSA(Bulked Segregant Analysis).Validation analysis of the marker was performed using F1 segregation population tested and resistant and susceptible genotypes generally known. Marker CCR-110 was found to show good resistance relevancy. Co-segregation analysis showed that correlation coefficient between the marker and citrus canker resistance was 0.79. The recombination frequency was calculated with maximum likelihood method
    and transformed into centimorgans(cM) according to the Kosambi function. The recombination frequency is 9.38%±3.5% and genetic distance is 9.11 cM. The marker was not found in susceptible genotypes (sweet orange, pummelo and trifoliate orange), but it was observed in most resistant ones (Kumquat,
    Ichang Papeda, Yuzu, and mandarin).

  • The fused pea ts-rbcS fragment and crtB gene from Erwinia herbicola was inserted into the PMV plasmid vector, resulting the plant expression vector pBI-ts-rbcS-crtB,which was transformed into tomato by Agrobacterium-mediated transformation. The result of PCR,Southern blot and RT-PCR revealed that foreign crtB gene was integrated and expressed in the transgenic tomato lines. Total carotenoids contents in the fruits of transgenic lines were 1.3–2.5 times higher than that of the control, the phytoene, lycopene,β-carotene and α-carotene contents were 4.3,1.8,2.2,and 2.3 times higer respectively.In addition,the expression of endogenous genes involved in carotenoid synthesis was affected in the transgenic lines. These results suggested that overexpression of crtB significantly enhanced the carotenoids biosynthesis and accumulation in transgenic tomato fruits.

  • The effects of air relative humidity(RH)on the photosynthesis and fruit-set of tomato seedling under high temperature were investigated in growth chambers. Three air relative humidity treatments from 10:00 to 16:00 were 70% ± 5%(high RH),55% ± 5%(middle RH)and non-humidification
    (low RH,as control). The results showed under the high temperature(the average temperature from 10:00 to 16:00 was 33–43 ℃),high RH treatment significantly promoted transpiration rate and net photosynthetic rate,also decreased or eliminated the photosynthetic midday depression compared with low RH treatment.Hence,the cumulative value of transpiration and photosynthesis from 9:00 to 17:00 of high RH treatment were 116%–133% and 31%–343% higher than that of low RH treatment,respectively. The
    fruit-set rate of high RH treatment was up to 48% and significantly higher than low RH treatment. However, photosynthesis and fruit-set rate of middle RH treatment were not significantly different compared with that of low RH treatment. As a conclusion,relative humidity of 70%(high RH)improved
    photosynthesis and fruit-set of tomato under high temperature above 30 ℃.

  • The 67 squash samples with viral symptoms collected from seven squash-growing counties in Chongqing were detected by antigen–coated plate (ACP)-ELISA with antibodies against seven plant viruses. The results showed that at least 59 samples with in these 67 squash samples were infected by the
    viruses, of which 80.60%、77.61%、70.15%、82.09%、76.12%、83.58% and 55.22% of the samples were infected by Cucumber mosaic virus (CMV), Tobacco mosaic virus (TMV), Tomato mosaic virus (ToMV), Turnip mosaic virus (TuMV), Broad bean wilt virus 2 (BBWV-2), Potato virus Y (PVY) and Potato virus X (PVX), respectively. Among 59 positive samples, 93.22% of the samples were mix–infected with more than two viruses, and about 60% samples were mix-infected by the above seven viruses, indicating the mix406 infections of viruses on squash were severely. Moreover, the results showed that among 54 amples infected by CMV,49(90.74%)samples were infected by CMV subgroupⅠ(CMVⅠ)and 12(22.22%)samples by CMV subgroup. Comparison of the nucleotide sequences showed that it has the highest identity(98.1%) with the previously reported CMVⅠisolate infecting Phaseolus angularis(CMV-RB)from China.

  • Cloning,Characterization and Expression of a H+-PPase Gene CuPPase from Cucumis sativus L.

  • Using PCR degenerate primers designed based on the conserved amino acid sequences of known tonoplast H+-PPase to amplify cDNA fragments from cucumber by RT-PCR and RACE-PCR, a H+-PPase gene named CuPPase was cloned. The CuPPase protein showed 94%, 92% similarity to the H+-PPases from Cucurbita moschata and Vigna radiata. Bioinformatics analysis indicated that the full-length cDNA sequence was 2 650 bp, which contained an open reading frame of 2 307 bp and encoded a protein of 768 amino acid residues with a calculated molecular weight of 80 kD and isoelectric point of 5.32. There was fourteen strong inside to outside transmembrane helix. PlantCare analysis indicated that there were abscisic acid, somatotropic hormone, salicylic acid cis-acting responsive elements and gibberellin and low temperature,drought wound-responsive elements in the protein encoded by CuPPase. The result of RT-PCR analysis indicated that CuPPase expressions were different in roots,stems and leaves,and richer in leaves and roots, poorer in stems. The expression of CuPPase is induced by salt stress and low or high temperature stress, but is closely related to the processing time. Its expression trend increased firstly and then decreased.

  • The expression of disease resistance related genes in cucumber(Cucumis Sativus L.)seedlings inoculated by Cladosporium cucumernium strain with weak virulence was analyzed among 72 hours by cDNA-AFLP method in this study. The results showed that the difference of gene expression began at 3 hours after inoculation, and the number of differential expression fragments reached the maximum at 6 hours after inoculation. The number of induced gene fragments was more than that of inhibited gene fragments at 8 time points after inoculation, but the results was not the same at 12 hours after inoculation. Through recovery,cloning,sequencing and homology analysis,5 obtained fragments share high homology with salicylic acid lucosyltransferase,Pyrus communis phosphatidyl glycerol specific phospholipase C-like mRNA,etc. respectively. We concluded that disease resistance of cucumber induced by inoculation of Cladosporium cucumernium strain with weak virulence might depend on signal transduction pathway mediated by salicylic acid,further induce system acquired resistance(SAR). Phospholipase C,as one of the key enzyme of signa transduction in stress tolerance,could be involved in the process described above. Northern blot results further proved the differential fragments was induced by the inoculation of Cladosporium cucumernium.

  • Effects of ABA on the Development and Dormancy in the Bulblets of Oriental Hybrids Lily ‘Siberia’ Generated in Vitro

  • The effects of abscisic acid (ABA) on the development of morphology and dormancy of bulblets in vitro of Oriental hybrids‘Siberia’were investigated. The results showed that the primordium was developed into scales only in the dark. While Fluridone, an inhibitor of ABA-synthesis, was applied during in vitro culture, the primordium was developed into scales or leaves. While exogenous ABA applied simultaneously with Fluridone, the effect of Fluridone was nullified. It indicated that the exogenous ABA can inhibite the formation of scal-leaves while endogenous ABA-synthesis was inhibited. Fluridone can reduce the levels of dormancy, and the function can be reversed while adding exogenous ABA. It
    suggested that the exogenous ABA can promote the development of dormancy. The dormancy levels of in vitro bulblets cultured at 15, 20 and 25 ℃ will be raised , but the low concentration exogenous ABA had no effect on the bulblets at 15, 20 and 25 ℃ with a low level of dormancy. It speculates that the effect of temperature on the dormancy maybe not play by ABA, an unknown factor , in addition to ABA, still played a key role in dormancy development. There is no relation between the morphology development
    and the dormancy levels.

  • A all-length heat shock transcription factor(HSF) gene with 350aa ORF, belonging to HSFA, was cloned from Lilium longiforum ‘white heaven’ by RACE(Rapid Amplification of cDNA Ends). Cluster analysis showed that this gene was highly homologous to known HSFA2 from other organisms and one novel member of HSFA2. Furthermore, alignment with HSFA2 from tomato and Arabidopsis indicated there was highly conserved DNA binding domain in this gene, harboring other characterized domains and regulatory elements. RT-PCR showed that expression of this gene could be detected in roots and bulbs
    treated by 37 ℃ for 1 hour and be detected in leaves during 0.5–12 hours at 37 ℃.

  • FT gene was cloned by RT-PCR from wild type Arabidopsis. After sequencing, the FT gene with proper digestion sites was digested and inserted into plant expression vector Super1300+, and was introduced into Chrysanthemum genome by Agrobacterium-mediated transformation. The integration of
    FT was determined using PCR and PCR-Southern, and gene expression was tested by RT-PCR. After transformation, we obtained 29 resistant plants. PCR and PCR-Southern blot analysis showed that eight of them were positive. RT-PCR detected the expression of the exogenous FT gene in the leaves of transgenic plants. One line of eight transplants appeared flowering bud in vitro. It indicated that transplants flowered early without influence of light.

  • Vasing physicological characteristics of cut peony flowers in various development stages and the regulating effect of fresh-keeping solution on the quality and vase life of cut flower were studied. The results showed that the vase life and the optimum ornamental period of cut peony flowers are
    relatively short under distilled water (control) with the flower stage increasing from bud occuring color stage to full-blooming stage, whereas were prolonged under the floral preservative which could raise the max flower diameter and the fresh weight of flowering branches. In vase period, the water absorption and loss of flowering branches decreased continuously, and the water balance value reached the maximum at 1 day of inserting in distilled water condition. However, fresh-keeping solution could improve water status, and delay the time of hydrologic balance value going down to zero, which showed significantly positive correlation with vase life. It is therefore proposed that the optimum harvesting period of cut peony flowers is from soft bud stage to begin opening stage, and the floral preservative could improve the quality of cut
    peony flowers in vase-holding.

  • The development process of spores of Adiantum flabellulatum in sterile culture condition and effects of different factors on spore germination and gametophyte development of A. flabellulatum were studied. The results showed that the spore germination was of the Vittaria-type and the gametophyte
    development was of the Adiantum-type. Spore could not geminate in darkness. Proper time of deepfreeze was good for spore germination rate. After 20 days of spore sowing, the maximum germination rate was obtained in 1/4MS medium, which reached 22.7%; additional 15–45 g · L-1 sucrose in culture medium were benefit spore germination,and sucrose was beneficial to gametophyte sexual differentiation;the sporophyte formation rate could reached 48.3% in MS + 30 g · L-1 medium;additional auxins were not beneficial to sporophyte formation,but additional NAA 0.5 mg · L-1 could promote gametophyte multiplication;sporophyte accommodation in sterile sand bottles could support a high alive rate about 90%.

  • Studies on Genetic Background of Important Germplasm Resources Among Citrus Based on cpSSR and EST-SSR Marker

  • Chloroplast simple sequence repeat(cpSSR)and expressed sequence tag-simple sequence repeat(EST-SSR)markers were used to reveal Citrus and its related genera phylogenetic relationship and to clarify the genetic background of some unique citrus germplasms conservered in National Citrus Germplasm Repository(Chongqing). Fifty and forty-four accessions were used for cpSSR and EST-SSR analysis respectively. Twenty-two pairs of cpSSR specific primers generated 146 bands with an average of 6.64 alleles per locus and an average Shannon’s information index of 0.32;Eighteen pairs of EST-SSR primers produced 240 bands with the mean of 13.3 alleles per locus,the mean polymorphic information content(PIC)was 0.73 and expected heterozygosity was 0.78. In addition, the genetic origin of some unique citrus accessions were also clearly clarified by cpSSR and EST-SSR analysis combining with the
    morphological characteristics,meanwhile,it was showed that Ichang Papeda have close phylogenetic relationship with Mangshan Yeju and Yuan Ju.

  • In order to analyze pistachio genetic relationship, random amplified polymorphic DNA (RAPD) was used to study the classification and identification of 13 pistachio (Pistacia vera L.)cultivars which includs 12 exotic varities and one native selects of Xinjiang. Twenty-one 10 bp primers that selected
    from eighty arbitrary primers were applied for amplifying the pistachio DNA. Total 137 bands were produced, among which 122 bands (89.05%) were polymorphic. Genetic distances among the cultivars were estimated based on the amount of band sharing and ranged from 0.2015–0.5163, it is indicated that there are genetic differences among the cultivars. UPGMA cluster analysis showed that 13 pistachio varieties can be divided into three groups in the genetic distance of 0.40 level: The first category includes eleven varieties, the second category is the ‘Xinjiang selection’ and the third category is ‘Mateur’ varieties.

  • This experiment was conducted to explore the effects of salt stress on stomatal movements and differentiation of amaranth leaves. Responses of stomata to different concentration of NaCl stress (0,50,150,300 and 500 mmol · L-1) , stomatal movements and differentiation of different phyllotaxy
    leaves after long time treated with 300 mmol · L-1 NaCl were investigated using the Amaranthus tricolor L. plant as materials. The results showed that the stomata of amaranth leaves closed rapidly after treated by salt. Stomatal aperature falled to the lowest after three-hour salt stress, afterwards, raised a litter. After 24 hour salt treating, most stomatas were closure except 50 mmol · L-1 NaCl treatment. Long time salt stress increased slightly stomatal density of the lower leaves, and reduced remarkably stomatal density of the upper fresh leaves. Long time salt stress also reduced stomatal aperature. Salt stress showed a more notable effect on stomata in the newer differentiation leaf.

  • Leaf,petiole and pedicle explants of Pulsatilla koreana were cultured on MS mediumsupplemented with various concentrations of zeatin,kinetin or 6-BA combined with 0.05 mg · L-1 IAA. After 6 weeks of culture,effects of cytokinin on adventitious shoot formation from explants were investigated. The highest frequency of shoot formation was obtained when petiole and pedicle explants
    were cultured on MS + 0.5 mg · L-1 zeatin + 0.05 mg · L-1 IAA. Regenerated shoot were cut as several cluster or single plant with callus like tissue, and then transferred on MS + 0.5 mg · L-1 zeatin + 0.05 mg · L-1 IAA or MS + 1.0 mg · L-1 6-BA + 0.05 mg · L-1 IAA for shoot propagation and growth. When shoot reached more than 3 cm in length, they were transferred on to root induction medium. 93 percent of rooting ratio was observed in 1/2 MS + 1.5 mg · L-1 NAA,after 42 days of culture. The rooted plantlets were acclimatized on mixture of North east mountain soil and perlite(2︰1).

  • Bentgrass is a major cool season turfgrass. Few molecular markers have been developed for bentgrass compared to field crops. In this study, 1 058 EST-SSR primer pairs developed by our lab from wheat, barley, maize, sorghum and rice were screened on bentgrass. Of which, 620(58.60%) primer pairs showed amplification. Effective primer pairs from wheat, barley, maize, sorghum and rice were 313 (61.49%), 30 (63.83%),101 (51.53%),11 (35.48%),and 165 (60.00%), respectively. Furthermore, 53 EST-SSR primers were selected to investigate the genetic relationship of eight bentgrass cultivars, and 51
    (94.34%) primer pairs showed polymorphism. In total, 368 alleles were found with an average of 2.69 alleles per primer. The PIC (polymorphism information content) for each polymorphic primer varied from 0.49 to 0.93, with an average of 0.78. The average genetic similarity coefficient (GS) among the eight cultivars was 0.72(ranged from 0.48 to 0.86). Clustering analysis indicated that the eight bentgrass cultivars were clustered into two groups based on 0.75 of GS. GroupⅠconsists only Highland, group Ⅱincludes the others, which could be divided into two subgroups based on 0.80 includes Alpha, Penncross and Blackspin, The sub-group Ⅱ-2 includes Putter, L-93, Cato and Penn A4.

  • ‘Zaofusu’ is a new pear cultivar with extremely early maturity which is selected from the crossed progeny of Pyrus bretshneideri ‘Dangshan Suli’ and P. communis ‘Beurre Giffard’. The average fruit weight is 147.8 g. The content of soluble solids, soluble sugar, total acid and vitamin C is 12.3%, 8.9 %, 0.31 % and 143.0 mg · kg-1, respectively. The fruit core is small, flesh is juicy and delicious. It has a little sclereid. The fruit matures in the early July in the area of Dangshan County, Anhui Province.Its development period is less than 90 d. The cultivar is resistant to Psylla chinensis and alkali-saline soil. It is
    suitable to be cultivated in the area of the Ancient Yellow River Valley and South China.

  • ‘Fucuimi’is a new Zizyphus jujuba cultivar which selected from clones of‘Zaozhuang Cuizao’. It matures about 15–20 d earlier than that of‘Zaozhuang Cuizao’. The fruit is 1.86 times as heavy as ‘Zaozhuang Cuizao’, the total sugar content has been increased by 5.7% compared with
    ‘Zaozhuang Cuizao’. It has red color, good flavor, excellent quality and strong adaptability. Trail results proved that ‘Fucuimi’is superior to‘Zaozhuang Cuizao’and the other cultivars grown in North China. The cultivar has high and stable production, which should be popularized over large areas.

  • ‘Ji P135’and‘Ji S136’were two new tomato hybrids developed by crossing lines ‘JBF2-2-3-M’with‘HZF2-3-1-M’,‘FGF2-1-3-M’with‘HZF2-3-3-M’. Main characters of ‘Ji P135’are as follows: early maturity, round fruit, hard fruit, thick skin, about 230 g per single fruit weight, output was 90 000–97 500 kg · hm-2. Characters of‘Ji P136’are as follows: mid-early maturity,round fruit without green shoulder,beautiful shape,good quality,high fruit setting rate,about 250 g per single fruit weight,output was 97 500–105 000 kg · hm-2. Both two tomato hybrids have moderate growth power and sparse leaves,ventilation and its light transmission are good while it are easy to plant and manage.

  • ‘Xinxiang Lajiao 4’is a new hybrid cultivar developed by crossing female parent ‘C-98-1’with male parent‘98-160’. The hot pepper has long sheep-horn fruit shape,20–25 cm fruit length,4–5 cm fruit diameter,0.35 cm pulp thickness and 50–70 g fruit mass. The cultivar also has many great charactors,such as beautiful light green color,straight fruit-shape, spicy flavor, prematurity and resistance to bacterial wilt,Phytophthora capsici,as well as virus disease. Its yield is up to 67 500–90 000 kg · hm-2. It is suitable for cultivation in protected field in the northern and open field in the southern.

  • ‘Jiza 1’is a new cucumber hybrid developed by crossing inbred lines 96-12-3-8-2-3 with 96-8-9-2-6-4. The cultivar has excellent characteristics such as early maturity, vigorous growth, resistance to downy mildew, fusarium wilt and angular leaf spot. The fruit is dark green, tender, crisp and it is about 28–33 cm long, 200 g weight. Its yield is up to 90–105 t · hm-2. It is suitable to cultivation in plastic tunnel in spring and autumn.
