  • Histocytology study was done by paraffin section method on the globular embryogenesis in pineapple(Ananas comosus). The results showed that the non-embryonic callus were cultured on the MS medium contained 4.0 mg · L-1 2,4-D and 0.5 mg · L-1 6-BA, embryogenic cells appeared in it. Then the single embryogenic cells formed a proembryo composed of many cells through cell division. The cells of proembryo had strong division ability, and there were distinct difference in shape and obvious boundary between them/it and the surrounding tissues. A suspensor-like structure was formed on the bottom of the proembryo. Along with the constant growth of the proembryo, its shape converted from long-ellipse to obovate, at last it evolved as a globular embryo with smooth surface, and there were absciss layer in the joint of the bottom of suspensor and matrix tissue. At the same time, cell differentiation appeared in the globular embryo. Pineapple somatic embryogenesis has two original modes,endogenous origin and exogenous origin. Additionally, there were ubiquitous uneven,desynchronization and secondary embryosphenomenon during somatic embryogenesis in pineapple callus.

  • A high-resolution genetic linkage map of litchi(Litchi chinensis Sonn.)was constructed from a compatible mating between isolates from the ‘Maguili’ and the‘Jiaohe Sanyuehong’groups with Random-Amplified Polymorphic DNA(RAPD),Sequence-Related Amplified Polymorphism(SRAP) and Amplified Fragment Length Polymorphism(AFLP). The linkage analysis produced 20 linkage groups in ‘Maguili’,which contained 238 markers,covering a total genetic distance of 1 096.59 cM. The physical size to genetic distance was approximately 4.61 cM;Nineteen linkage groups were regenerated in‘Jiaohe Sanyuehong’,which contained 239 markers, covering 881.36 cM,with an average genetic distance of 3.69 cM

  • A UCE gene, encoding ubiquitin conjugating enzyme was isolated from banana fruit(Musa acuminate L. AAA group‘Brazilian’)and designated MaUCE1. The full length of MaUCE1 cDNA is 890 bp encoding 152 amino acids that shared high identities with the UCE from Nicotiana tabacum, Triticum aestivum, Solanum tuberosum, Arabidopsis thaliana(95.39%, 95.39%, 92.11% and 90.79%). Amino acid alignment indicated MaUCE1 exists an enzyme activity site of E2 and a conserved cysteine, which is considered to catalyze formation of thioester bond. RT-PCR analysis showed that MaUCE1 is ubiquitously expressed in roots, stems, leaves, flowers and fruits, but the expression level is higher in fruit and flower than those of other organs. During the process of banana fruit postharvest ripening, the mRNA accumulation of MaUCE1 increased in evening of fruit ripening, which is in line with the change of starch phosphorylase activity but negative correlated with change of starch content in this period, suggesting thatMaUCE1 might involve in the catabolism during the process of banana ripening,rather than trigger the fruit ripening.

  • A green fluorescent protein (sGFP) tagged Foc4 strain was developed by the method of PEG-CaCl2. The fluorescent signal was clearly visible in cytoplasm and nucleus of the hyphae and spores under the fluorescence microscopy. The transformed Foc retained its pathogenicity and growth patterns. Water culture of 9 banana cultivars with different resistance were performed for 15 days using 200 mL of sterile distilled water for each plantlet,and the root exudates were collected and bioassay for their effects on the GFP tagged Foc4. The results showed that the apolar fraction of the root exudates from the highly resistant cultivars could kill the pathogen. Those of the general resistant cultivars could inhibit the spore germination and hyphal growth,while the sensitive cultivars couldn’t.

  • Fragan × ananassa Duch.‘Toyonoka’plants were exposed to 5,10 and 15 mg · L-1 cadmium(CdCl2 · 2.5H2O)for 6 d to investigate the influence of heavy-metal Cd on root tips,which included DNA polymorphism change and oxidation systems damage. In addition,above indexes were analyzed by physiology biochemistry methods and random amplified polymorphic DNA(RAPD)technology. Cd induced an inhibited effect on root growth and soluble protein content,which also accumulated reactive oxygen species outbreak including superoxide anion production rate increased and hydrogen peroxide content enhanced. At the same time,antioxidant enzymes(SOD,POD and CAT)activities also decreased. Moreover,DNA hyperchromic effect reduced after 6 d on 5,10 or 15 mg · L-1 Cd treatment. Finally,RAPD results showed that 8 primers gave a total of 88 RAPD bands ranging from 150 to 3 500 bp in molecular size in the normal plantlets. The changes occurring in RAPD profiles of the root tips after Cd treatment, which included loss of normal bands,appearance of new bands and variation in band intensity in comparison to which of the plantlets without treatment. It is need to point out that the RAPD band change effect was dose-dependent of Cd concentration. These results indicated that genomic template stability(a qualitative measure reflecting changes in RAPD profiles)was significantly affected by the above Cd concentrations. Reactive oxygen species outbreak and DNA cross-linking were the main reasons which causing the root tips damage. Thus,DNA polymorphisms detected by RAPD analysis could be used as an effective method for the assessment of the root tips damage by Cd in strawberry plantlets.

  • Effects of Exogenous Calcium on K+,Ca2+,Mg2+ Content and ATPase Activity in Cucumber Seedlings Under Root-zone Hypoxic Stress

  • A hydroponic experiment was carried out to explore the effects of exogenous calcium and H+ on ion content and ATPase activity of cucumber under hypoxic stress. The results showed that K+,Ca2+,Mg2+ contents were significantly lower and plasma membrane,tonoplast,endoplasmic reticulum ATPase activities were significantly decreased under hypoxic stress. Application of 4 mmol · L-1 CaCl2 in nutrient solution significantly alleviated the hypoxic stress,and the K+,Ca2+,Mg2+ contents were significantly higher than those in the hypoxic treatment,while the plasma membrane,tonoplast,endoplasmic reticulum H+-ATPase and Ca2+-ATPase activities were significantly higher than hypoxic treatment and closed to the control level. Application of 50 μmol · L-1 H+ in nutrient solution significantly reduced the contents of K+,Ca2+,while the content of Mg2+ was not significantly differed from the hypoxic treatment. H+-ATPaseand Ca2+-ATPase activities of plasma membrane were significantly lower than the hypoxic treatment,but the H+-ATPase and Ca2+-ATPase activities of vacuole membrane and endoplasmic reticulum were not significantly differed from the hypoxic treatment. It is suggested that application of exogenous calcium ions can increase the mineral absorption and transport,enhanced the activities of ATPase,and alleviated the hypoxic injury of cucumber and enhanced cucumber tolerance hypoxia.

  • In order to study effects of salt stress on changes of soil microbial quantities and the diversity of soil microbial community by the technology of PCR-DGGE, two kinds of cucumbers with different saline tolerance were selected,Jinchun 5(salt-tolerant)and Jinyou 1(salt-sensitive),and four concentrations of NaCl were designed(0,100,200 and 300 mmol · L-1). The results showed that salt stress could reduce the richness of soil bacterial community significantly(P<0.05). The richness of salt-sensitive cultivar was high under low salt treatment, while the richness of salt-tolerant cultivar was high under high salt treatment. High salt stress had more effects on the rhizosphere bacterial community structure of salt-tolerant cultivar. Sequences of different bands of four stages belonged to four bacterial groups,Firmicutes,Proteobacteria,Gemmatimonadetes and Bacteroidetes. Most of them belonged to Alphaproteobacteria and Gammaproteobacteria. With the increasing of salt concentration,the plant heights of cucumber and yield per plant reduced.

  • The effects of cytokinin(CTK)on growth, optical absorption and chlorophyll fluorescence kinetics parameters of ovary and fruit were studied in tomato ‘Bobcat’. A floral bud at the 7 d before flowering or a fruit at the 10 d after flowering was respectively sprayed with 600 and 800 mg · L-1 CTK. The CTK significantly promoted expansion of ovary and fruit. However, the vertical length of ovary of spraying the floral bud with 600 mg · L-1 CTK was obviously superior to 800 mg · L-1 CTK, and vertical length of fruit of spraying the fruit with 800 mg · L-1 CTK was obviously superior to 600 mg · L-1 CTK. The absorption coefficient(Abs)on the surface of the ovary could be in higher levers with 600 mg · L-1 CTK than 800 mg · L-1 CTK, but the Abs on the surface of the fruit could be in higher levels with 800mg · L-1 CTK than 600 mg · L-1 CTK. With the development of ovary and fruit, the maximum quantum yield of primary photochemistry of photosystem Ⅱ(Fv/Fm), original quantum efficiencies(Q)and maximum electron transfer rat (ETRmax)decreased gradually,while the non-photochemical quench(qN) increased gradually. The CTK increased light absorption and utilization of ovary and fruit, which promoted the growth and development of fruit.

  • To identify molecular markers tightly linked to the male sterility gene Ms in Chinese cabbage, a BC1 population was developed from the cross between male sterile plants(MsMs)and fertile double haploid line ‘FH-1’(msms). AFLP primers were screened for polymorphisms between the parents and two bulks representing fertile and sterile plants. Out of 512 primer combinations, 3 combinations gave polymorphic amplification patterns. These polymorphic amplicons were cloned and successfully converted to dominant SCAR markers, designated syau_scr02, syau_scr03 and syau_scr05. All the SCAR markers are located on one side of the Ms locus with a map distance ranging from 1.60 to 2.50 cM. These SCAR markers show high polymorphism between ‘AB01’ and 12 fertile inbred lines.

  • In order to explore the effects of different inducers in enhancing the resistance against lily cut flower disease,roots and scales of lily bulb were soaked in chemicals such as DL-β-aminobutyric acid(BABA),Isolane(10% isoprothiolane,10% hymexazol),epibrassino1ide(epi-BR),hydrosoluble chitosan (OS),acibenzolar(BTH),abscisic acid(ABA)、MgO and NH4Cl before planted or embedded. Growth indexes,the activities of defensive enzymes such as POD,PAL,PPO and disease index were determined by three separated trials,those were potted test,field test and embedded scales test. The results showed that all the inducers involved in the research can increase activities of defensive enzymes and budding rate of scales,improve growth properties including the height,the number of the leaves and the
    thickness of stems,furthermore reduce disease. Compared with the control,the treatments with OS 400 significantly promoted plant physiological and biochemical indexes and disease resistance.

  • A FT-like gene named CmFT(Genbank accession No. GQ925916)was cloned by RT-PCR and rapid amplification of cDNA ends(RACE) using Chrysanthemum morifolium Ramat.. A predicated 525 bp open reading frame was deduced in the cloned sequence,which encoded 174 amino acids. By alignment analysis,the deduced amino acid sequence contained two critical residues and conserved region,both of which identified as diagnostic of true FT. The gene was proposed to belong to FT subfamily by phylogenetic analysis. Fluorescence quantitative RT-PCR system followed by SYBR GREEN was established to examine the expression of CmFT under different part of mature tissues and different photoperiod treatments. CmFT gene expressed highest in flower buds,next in stem and lowest in leaf. Expression of CmFT under the short day(SD)treatment was significantly high than that under long day(LD). Abundance of CmFT indicated a diurnal rhythm in SD-grown plants,so that its levels showed a decreasing trend under light,a rise during night and the maximal level before the coming of light. Under
    the LD treatment,the expression level tended to be zero. The results suggest that CmFT is associated with
    photoperiod sensitivity and may function in regulating photoperiodic flowering in Chrysanthemum.

  • The changes of color at different parts of the ligulate corolla and among the different whorls of the ligulate florets of red-,yellow- and white-flowered chrysanthemum cultivars were analyzed. It was found that the color of the medium part of the corolla of each ligulate floret is representative for the whole ligulate corolla,and the ligulate florets in the middle whorl are representative for the whole capitulum. The 120 chrysanthemum cultivars were randomly selected and measured the lightness L* and chromatic components a* and b*. The values of lightness L* and chromatic components a* and b* of the obverse side of the ligulate florets are closely positively correlated to the values of the reverse sides,and the ranges of the values of the obverse side are greater than those of the reverse side. It is thus better to use the color value of the medium part of the obverse side of the ligulate floret to define the flower color accurately. Analysis of the three-dimensional scatter plots revealed some chrysanthemum cultivars as having rare flower colors,which constitute important resources for breeding chrysanthemum with novel flower colors.

  • Taking terminal buds of different developmental stage of Tagetes erecta L.‘Scarletade’as materials,the flower bud differentiation process,micro structure and ultra-micro structure of flower organs were studied by using morphological observation,paraffin section and SEM and TEM. The results showed that:(1)The process of flower bud differentiation starts from the two-leaf period to three-leaf period,and from outward to inward,the developmental stage is closely related to morphological changes.(2)The structure of the stigma of the flower changes from bar-like to Y-type and finally to curved Y-type,there are plenty of papilla structures covered the surface,the stigma became curved Y-type after pollination and wizened papilla structures.(3)There are two pollen sacs in mature anthers,which were separated by connective tissues. The mature pollen sac wall is composed of flat epidermal cells and fibrous layer.(4)The pollen are prolate,ellipsoidal in equatorial view and nearly spherical in polar view,with 3 germinal furrows,and there are many spines of different length,with the average of 4.65 μm,on the exine. The equatorial axis length is 37.5 μm,and polar axis length is 25.5 μm.

  • Different levels of Na2SeO3(0,50,100,200 and 400 μg · mL-1) and(CH3COO)2 Zn · 2H2O(0,0.2%,0.4%,0.8% and 1.2%) was applied by foliar spraying to determine the effects of Se,Zn and the their interaction on the subcellular distribution of Selenium in spring tea leaves. The result showed that:(1)The Se content in subcellular part of the cell wall,membrane and organelle part,soluble fraction compared with the control increased in varying degrees in different treatments of Se. The Se was mainly distributed in membranes and organelles.(2)The increased effects of Se content in different subcellular components was obvious in lower concentration(0.2%–0.4%)of zinc treatment,but the Se content of the different subcellular components was significantly lower and the Se assimilation was restrained when the zinc concentration increased to 0.8%–1.2%. (3)The content and the distributing proportion of selenium in membrane and organelle part were the highest,and the second was cell wall,the third was soluble fraction. Combined applying middle concentration of Zn(0.4%–0.8%)and Se(100–400 μg · mL-1) increased obviously the Se content in different subcellular component,but combined applying high-concentration of Se and different Zn concentrations had no significant effects on the Se accumulation. The Se in cell wall and soluble fraction was transferred to membrane and organelle part obviously when combined applying lower Se concentration(50–100 μg · mL-1)and different Zn concentrations. The Se of subcellular was transferred mainly in the between the membrane and organelle part and soluble fraction when combined applying higher Se concentration and different Zn concentrations. In general,whether single Se and Zn treatment or Se-Ze combined applying treatment,it could significantly affect the Se distribution in tea subcellular cells. The Se was mainly distributed in membranes and organelles by Se and Se-Zn interaction,and accumulated in cell wall by single Zn treatment.

  • With Hamburg grape mesocarp from berries during 7–11 weeks after flowering, the expression levels of VvNCED1 and VvBG1 gene were detected through real-time quantitative PCR. The results showed that the expression level of VvNCED1 gene was very high during 7–8 weeks after flowering, followed by a rapid decline to its lowest point, and then increased a little. Coupled with the onset of berry ripening and coloring-up, the mRNA expression of VvBG1 gene, on the whole, increased continually. Given that the ABA continual rising during the onset of grape berry ripening, it is considered that, the ABA accumulation is attributed to high mRNA expression level of VvNCED1 before the onset of berry ripening and high mRNA expression level of VvBG1 after the onset of berry ripening.

  • The effect of moisture content of medium on transpiration, fresh matter accumulation and related physiological indexes for potted tomato in greenhouse were studied through setting water supply tension at 10,30,50 and 70 hPa using negative pressure pot device. The results indicate:The moisture content of medium can be accurately controlled and maintained by the negative pressure pot device,moisture contents of medium are 88%,76%,63% and 57% at water supply tension 10,30,50 and 70 hPa correspondingly. In different experimental stages,influence of moisture content of medium on daily transpiration is different. Accumulative daily transpiration of potted tomato is as following:10 hPa treatment>30 hPa treatment>70 hPa treatment>50 hPa treatment. The effect of medium moisture content on fresh matter accumulation is the most significant at 30 hPa,followed by 10 and 50 hPa and the least is at 70 hPa. The effect of water supply tension on yield, water consumption and crop water productivity are extremely significant. 30 hPa is the most favorable water supply tension for potted tomato in greenhouse because of the maximum yield and the fresh matter accumulation.

  • Lilium brownii var. viridulum Baker is an important medicinal plant species. Here we employed inter-simple sequence repeat (ISSR) markers to explore genetic diversity and patterns of genetic variation in 100 individuals from four cultivated populations in Jiangxi Province. Using 14 ISSR primers, 213 discernible DNA fragments were scored with 165(77.5%)being polymorphic. High levels of genetic variation were revealed both at the population level(mean PPF = 31.69%,Ne = 1.198,I = 0.168,Hpop = 0.113)and the species level(PPF = 77.46%,Ne = 1.438,I = 0.392,Hpop = 0.260). Strong genetic differentiation among populations(AMOVA:FST = 0.632,P < 0.001)was also detected, which may result from artifical selection and long-term cultivation. The UPGMA cluster analysis and principal components analysis (PCA) revealed that individuals from the same population formed a distinct group,and LS population was clearly separated from other populations. On the basis of these observations, some suggestions for efficient management of the genetic resources of this important medicinal herb are proposed.

  • Aromatic components in flower sample of Paeonia lactiflora ‘Yangfei Chuyu’ and P. lactiflora ‘Dafugui’ were determined using the methods of headspace solid-phase microextraction(HS-SPME)and gas chromatography-mass spectroscopy(GC–MS). All sorts of aromatic compounds and their relative content at different stages in opening progress were compared and analyzed. 34 kinds of aromatic compounds were consequently detected in ‘Yangfei Chuyu’ sample and 36 kinds in ‘Dafugui’ sample. For ‘Yangfei Chuyu’,nerol,myrcene,α-pinene,3-carene and spinacen comprised the principal aromatic compounds;While for ‘Dafugui’,phenylethyl alcohol,3-hexen-1-ol,hexyl alcohol,diethyl phthalate and 1,4-dimethoxy-benzene to be principal.

  • The effects of three plant growth retardants paclobutrazol(PP333),uniconazole(S3307) and chlorcholinchlorid(CCC) and two methods of application(foliar spray and root drench) were studied on Zamioculcasz amiifolia plants of ages one,two and three years. PP333 and S3307 both significantly inhibited growth and their effects increased as the concentration applied increased. However,CCC did not significantly inhibit growth. PP333 reduced the average total length of leaf stalks,minimized the length petiolules(i.e. the stalk between leaflets);shortened and thickened the petioles and also reduced the comparative growth speed of compound leaves. The optimal concentration of PP333 at different ages of Z. amiifolia varied with individual plant biomass. In contrast,the main effects of S3307 were on shortening and thickening petioles. Overall,the best control growth control of one-year-old Z. amiifolia plants was obtained with 300 mg · L-1 of S3307 applied as a foliar spray;of two-year-old plants with 300 mg · L-1 of S3307 or 600 mg · L-1 of PP333 applied as root drench;and of three-year-old plants with 1 200 mg · L-1 of PP333 applied as foliar spray.

  • Development of a Multiplex PCR Protocol for the Detection of Three Viruses in Banana

  • Based on the establishment of the optimal PCR and RT-PCR for the detection of single virus,a multiplex PCR protocol for the detection of three main viruses (Banana bunchy top virus,BBTV;Cucumber mosaic virus,CMV;Banana streak virus,BSV) in banana is developed. Three specific fragments with molecular weights at 615 bp for BBTV,480 bp for CMV and 945 bp for BSV were simultaneously amplified in one PCR reaction system. The sequence analysis of amplified fragments indicates that three virus fragments shared above 91 percent identities with that of other relative viruses. It was showed that the sensitivity of this protocol equaled to be able to detect the three viruses at the level of 10-2 mg banana tissue.

  • High molecular weight nuclear DNA(HMW-DNA)from leaves of Guanximiyou [Citrus grandis (L.) Osbeck ‘Guanximiyou’] was prepared by two methods(Zhang’s and Peterson’s method)and the extracting effects of the two preparing methods were compared. The results showed that Zhang’s method was more suitable for preparation of HMW-DNA in Guanximiyou. The cell walls were broken down by gentle physical homogenization to keep integrity of nuclei under suitable osmolarity. Debris of cell walls and most of polysaccharides were removed by filtering with miracloth and centrifugation under slow speed. The isolated nuclei were proved to be intact,pure,and high-quality by observation under microscope and were embedded with low-melting-point agarose to preserve the isolated nuclei in plugs for keeping stability of nuclei. The optimum digestion time of plugs was 48 hours,the pulse field gel electrophoresis (PFGE)results showed that the isolated HMW-DNAs were more than 1 Mb in size and were digested completely or partially by restriction enzymes,therefore,Zhang’s method was suitable for construction of BAC library in Guanximiyou.

  • ‘Hua gao’is a bud variation cultivar from‘Xingao’. The ripening date of pome is in the third ten days of August in Changsha area. The fruit is oblate with snuff color pericarp. The average weight of single fruit is 298.2 g,soluble solids content is 13.0%–14.1%,titratable acid content is 0.03%–0.10%,and vitamin C content is 52.5–65.0 μg · g-1. The multiple spots regional trials and tests showed that the cultivar has excellent characteristics such as high sugar,high yield,early bearing and good resistance for storage.

  • ‘Hechun’and‘Yongchun’are two new peach cultivars both for table and ornament,which were selected from G4 breeding by Institute of Agricultural Integrated Development,Beijing Academy of Agriculture and Forestry Sciences. Their flowers are double,pink and red in color respectively;corolla are large with 5.2 cm in flower diameter. The petal number range from 22 to 27.Their flesh is edible and sweet. The average single fruit weight are 131 g and 118 g respectively. Soluble solids content is 10.0%–11.2%. Flowering begins in middle and late April and lasts for 15 days in Beijing area.Date for fruit maturity is in middle and late July.‘Hechun’and‘Yongchun’are excellent edible ornamental plants when they are used in scenic spot,courtyard,park,orchard and vase art.

  • ‘Kuijin’apricot is a early maturing cultivar originated from a cross‘Honghebao’בErhuacao’. Its fruit shape is nearly round with an average fruit weight 89.1 g. The fruit skin is orange,fruit surface is bright,clean and beautiful. The flesh is yellow,fine meat,juicy,sour-sweet and aroma,with 4.28 kg · cm-2 fruit hardness,13.2% soluble solids content,10.9% total sugar content,0.93% titratable acid content. The cultivar has big fruit,good quality,high yield,multiple resistance,self-pollinated,early bearing and so on.

  • ‘Jing Ou 2’is a new middle-ripening Chinese dwarf cherry cultivar which is selected from seed seedlings in 1999,with the average single fruit weight 6.1 g,juice production 81.5%,the content of soluble solids 14.7%,soluble sugars 7.54%,total acids 1.32%,sugars and acids ratio 5.71,calcium 262 mg · kg-1,vitamin C 449 mg · kg-1,and total amino acids 5.22 g · kg-1. The fruits ripen in the last ten days of July in Beijing areas.

The size of nuts is middle,with flesh fine and smooth,and the nuts are perfect for stir-fry. The new cultivar‘Hongli 2’is developed aptly in the mountains and hills in Taiyi region,and at river bench area in Southeastern Shandong.

  • ‘Wanzao 1’is a new table cultivar of Chinese jujube(Ziziphus jujuba Mill.)selected from the local germplasm in Fuyang,Anhui Province. It ripes in early september in Hefei,Anhui,with 80–85 d fruit developing periods,super quality for fresh-consuming,high yield,and high tolerance to hardness. The fruit is round,with 17.04 g average mass,26.76 g maximal mass,98.0% edible rate. The flesh is crisp,succulent,delicate,and sweet.‘Wanzao 1’is a good disease tolerant cultivar and should be popularized over large areas.

  • ‘Nongda 601’is a new mid-early eggplant hybrid which has tight plant shapes and high constant fruit setting. The hybrid came from 03-621 × 03-151. The fruit is oblate-shaped and bright black, which has a few seed and a delicate flavour. The average single fruit mass is above 500 g . Its yield is up to 75–105 t · hm-2. It is suitable for protected cultivation and open field cultivation in spring.

  • ‘Hongxingyu’is a new seeded and small-sized watermelon cultivar which was developed from sexual hybridization. The whole growth period is about 94 days and the fruit maturing stage is 29–30 days. The mass of single melon is 2.2–2.4 kg. The fruit is oval and its skin is green with about 17 greenish black stripes on the surface. The center and side sugar contents are 11.85% and 9.10%,respectively.‘Hongxingyu’has the feature of little crude fiber,good quality,high sugar content,wet and draught tolerance.
