  • Preliminary Identification of Drought Resistance of Chinese Wild Vitis Speciesand Its Interspecific Hybrids

  • Under potting drought stress condition , drought resistance of eleven accessions of eight Chinese wild Vitis species and 107 F1 hybrids of V. yeshanesis ×V. riparia was identified based on the evaluation methods
    of degree of etiolating (DE) , relative water content (RWC) , and plasma membrane permeability ( PMP) in the leaves. The results showed that the accession of ‘Yanshan21’of V. yeshanesis was the highest resistant to the
    drought stress ; the accession of ‘Taishan21’of V. adsricta had middle resistance ; the accessions of ‘Anlin22’of V. adstricta , ‘Taishan211’of V. amurensis , ‘Jiangxi22’and ‘Pingli27’of V. romanetii , ‘Gansu291’
    and‘Nanzheng22’of V. piasezkii ,‘Weinan23’of V. quinquangularis , and‘Guangxi21’of V. pseudoreticule lata had lower resistance ; ‘Tangwei’of V. davidii is susceptible to the drought stress. There are demonstrated continuous genetic segregation of drought resistance in F1 progenies of V. yeshanesis ×V. riparia. Specifically , a few hybrids showed higher heredity than resistant parents in the tolerance to drought stress.

  • Study on the Berry Genetic Diversity and Evaluating Criteria of Some Quantitative Characters for Seedless Grape Germplasm

  • Investigation and statistical analysis were carried out on berry color , shape , mass and size of 94 varieties of seedless grape and 40 varieties of Kyoho type grape collected in National Grape Germplasm Repository Zhengzhou for three years. The results showed that there were profile genetic diversity in seedless grape. Variation coefficient both berry mass and berry size were much greater than that of Kyoho type grape. The average mass of
    seedless grape was in range from 1.0 to 5.0 g . Almost half of tested varieties had medium mass in 1.5 - 2.5 g ,The available description standards both domestic and international was considered to be not suitable for evaluating berry size and berry mass of seedless grape. Based on the result of the study , the new additional method for this purpose are suggested.

  • ‘Robertson Navel’orange fruits from five representative areas of three ecotypes in Sichuan province were used to study soluble sugar accumulation and their related enzymes during the fruit development , especially the changes in activities of sucrose phosphate synthase (SPS) and sucrose synthase (SS) . The results showed that SPS played an important role in sugar accumulation in fruit during the fruit inflating stage , while SS in
    the synthetic direction was the key enzyme during the fruit ripening stage. The activities of SPS and SS in the cleavage direction in fruit were similar between different habitats , whereas SS activity in the synthetic direction changed significantly in the different habitats. It was suggested that although SS in the cleavage and synthetic direction regulated the sugar transport and accumulation though fruit sink strength , SS in the synthetic direction was the dominant enzyme which led sugar different accumulation in different habitats.

  • A famous litchi (Litchi chinensis Sonn. ) cultivar‘Nuomici’was used to investigate the changes of color and the contents of anthocyanins and flavonoids in pericarp during chilling2injuried temperature storage. The
    browning index increased greatly after storing for 14 d at 0 ℃and developed irreversible chilling damage on the 21th day. The apparent color parameters a , L and C values decreased continuously with storage time , indicating that the peel had less red feature and became darker. Within 7 days (from 14 d to 21 d) stored at 0 ℃, the values of a , L and Cwent down by 27195 %, 33158 %and 58147 %, respectively. The contents of anthocyanin , flavonoids and total phenolic in pericarp at 0 ℃decreased , and on the 21th day they were 18.08 %, 35.99 % and 41.64 %, respectively , as those at harvest day. Cold storage at 3 ℃partially inhibited the changes of all these parameters , thereby delaying the browning progress. The color parameters of a , L and C value were significantly correlated with browning index and anthocyanins contents , respectively ( P < 0.05) . Colorimeter can be used as objective parameter to designate the browning degree of chilling injury.

  • Effects of relative soil water content on the changes of chlorophyll content , photosynthesis and the parameters of chlorophyll fluorescence in pistachio ( Pistacia vera L. ) leaves were investigated. The results were as follows : the content of Chl1a and Chl1b and the net photosynthetic rate in pistachio leaves were decreased while the content of carotenoid were enhanced by drought stress. The activity of PS Ⅱand the parameter of chlorophyll fluorescence in pistachio leaves such as conversion efficiency of primary light energy ( Fv/ Fm) of PS Ⅱ, the photochemical quenching coefficient (qP) , the Non2photochemical quenching coefficient (qN) and the quantum yield of PS Ⅱelectron transport (ΦPS Ⅱ) were decreased respectively when the relative soil water content is 20 %. The reasons of Pn decreased were stomatal and nonstomatal limitation respectively in light and severe drought stress condition. Under drought stress , It is a primary reason of net photosynthetic rate decreasing in pistachio leaves that the the quantum yield of PS Ⅱelectron transport (ΦPS Ⅱ) were reduced by soil drought and high irradiance.

  • Agronomic characters and biochemical matter contents in flower buds and leaves of the two CMS lines 9704A and 8214A , of their each maintainers and F1 hybrids for sterile line ×restorer and maintainer ×restorer in pepper were studied. Results showed that differences in plant height between the two kinds of hybrids reach significant level . POD activity in sterile linespflower buds and leaves is higher than that in maintainersp, still the activity in hybrids for sterile line ×restorer is significantly , even very significantly higher than that in hybrids for maintainer ×restorer. Sterile lines contain less free praline than maintainers in the flower buds , and the content in hybrids for sterile line ×restorer is significantly , even very significantly lower than that in hybrids for maintainer ×restorer. IAA content in sterile linespflower buds and leaves is lower than that in maintainersp. ABA content in sterile linespleaves is higher than that in maintainersp, but the vise versa in their flower buds. No obvious laws are found in the effect of the male sterile genes on content change of IAA , ABA and ZRs in hybrids yet .

  • Effect of transferring efficiency of Agrobacterium tumef aciens pH value 、concentration of AS added in co-cultivation medium , temperature during co2cultural stage and days for co-cultivation were studied through PCR
    amplification and observation of green fluorescent protein expression by used GFP as reporter gene on cotyledon of ‘Zhongjiao 5’and‘Xiangyan 10’. The results show that when pH value of Agrobacterium solution is 5.2 , transformation efficiency of pepper cotyledon incubated on co-cultivation medium added 200μmol·L -1 AS increased significantly under 23 ℃and 5 days dark co-cultivation. The rate of expression of green fluorescent protein are up to 40 % on calli of cotyledon of both‘Zhongjiao 5’and‘Xiangyan 10’.

  • The growth of cucumber was strongly affected by soil water content. Either leaf number or leaf area was significant different among the water treatments. At the beginning of water stress , the free protein and proline content increased in leaf , this trend disappeared at time prolonged when the soil moisture remained at the 50 %- 60 % soil capacity. The content of Spd (spermidine) and Put (putrescine) in leaf and root of the plant increased with the moisture decreasing in the soil , but there was no significant difference of Spm (spermine) among the three treatments. ABA content of cucumber leaf was high when the soil water content was low.

  • Semi-leafless pea is the mutant of common pea. Cross experiments between semi-leafless pea and common pea were carried out to find out heredity of semi-leafless gene. The results showed that semi-leafless is governed by a recessive gene af . It is linked with i (green cotyledon) and lf (low initial flowering node) on the first chromosome in pea , and the order of them is lf , af and i , i . e. af is between lf and i . Cross-over value between af and i is 6.80 % , and 36.74 % between af and lf . A new semi-leafless vegetable pea cultivar with super-high yield and high nutritional quality. ‘Xucai 1’was bred and selected. Its fresh pods taste delicious. Its extremely developed tendrils can be sold as top grade Longxu vegetable. The tender stems and leaves are also high quality vegetable. Significant value of af in pea breeding was conclusive.

  • The effects of antioxidant system on the ultradry seed of Brassica caulorapa Pasq. were investigated. Ultradry treatment did not cause any changes in seed germination percentage and vigor index in the seed of B .caulorapa. Further more , it improved the storability of the seed during 30 days of accelerated ageing. Ultradry treatment enhanced superoxide generation rate , but did not affect the level of malondialdehyde (MDA) and the activities of antioxidant enzymes such as superoxide dismutase (SOD) , catalase (CAT) , guaiacol peroxidase (GPX) and ascorbate peroxidase (APX) in B . caulorapa seed. After 10 days of ageing treatment at 50 ℃, both the superoxide generation rate and the MDA content were lower in ultradried seed than in the control . The content of ascorbic acid (AsA) was higher in ultradried seed than in the control . Once rehumidified and imbibed , the antioxidant enzymes in the ultradried seed played a very important role to scavenge the ultimate product of lipid peroxidation reduced during seed storage in order to maintain the seed germination ability. A more stable antioxidant system was maintained in ultradried seed , which should account for the high vigor of the ultradried seed.

  • Two hundred and one isolates of Phytophthora infestans on tomato from eighteen provinces , cities and autonomous regions were identified by using five differential tomato lines containing different resistance genes ,
    which were Ts19 , Ts33 , W1Va 700 , LA1033 and L3708. The result indicated that the eight different physiological races were found out . Race T1 and T1 ,2 were widely distributed in 13 and 11 provinces with high frequencies of
    28.8 % and 28.4 % respectively. Followed by T0 and T1 ,2 ,3 occurred in eight and seven provinces with frequencies of 1819 % and 819 % respectively. Then race T1 ,2 ,3 ,4 occurred in four provinces with 7 % frequency. The race T1 ,2 ,4 , T1 ,4 and T3 were narrowly distributed only in one or two provinces with 3.0 % , 2.5 % and 2.5 % respectively. This is the first report in China.

  • Denitrification Loss and N2O Emission from Nitrogen Fertilizer Applied to Veg2etable Field

  • A field experiment was conducted to measure denitrification loss and N2O emission from nitrogen fertilizer applied to a field grown with eggplants by soil core incubation-acetylene inhibition method. The results showed that the amount of N2O emitted from soil (without N fertilizer applied) was N 8.2 kg·hm-2 , while the amount of N2Oderived from applied N fertilizer was N 25.6 kg·hm- 2 (8.6 % of applied N) . The corresponding figures for denitrification losse were N 22.8 and N 15.1 kg·hm-2 (5.1 %of applied N) respectively. The denitrification loss and N2Oemission amounts of the treatment with N fertilizer applied were significantly higher than that of the treatment without N fertilizer applied ( P < 0.01) . These results also indicated that nitrification2denitrification was one of main approaches of nitrogen fertilizer losses in vegetable field. And there were significant relationships between denitrification loss and N2O emission and soil water content ( P < 0.01) .

  • The experiment of controlled2release fertilizers (CRFs) on growth and development of salvia plant in pot was conducted in comparison with common compound fertilizers (CCFs) . The contents of chlorophyll , number of leaves , leaf area and growth potential of the salvia treated by CRFs were significantly greater than those by CCFs.There were higher dry matter mass and flower mass of salvia treated with CRFs compared with those by CCFs. Furthermore , the treatment with CRFs distinctly boosted florescence flourish and extended view period , which promoted salvia growth , but also avoided damage to the plants caused by excess use of fertilizers. The optimum amount for fertilizer application on salvia in the 25 cm ×21 cm pot was N 0.4 g/ kg soil .

  • The experiments were made with Dendrobium huoshanense. It was studied that photosynthesis and chlorophyll fluorescence of D. huoshanense. The results indicated that there were positive correlation between daily
    change of photosynthesis and it of light intensity under cultivation environment of forest shadow , as well as photosynthesis rate was low. Low light intensity is the reason to result in low growth of D. huoshanense and is not suitable
    light intensity for dendrobium growth in forest shadow. Daily change of photosynthesis represents V type and there is the lowest photosynthesis at noon under natural light . From 9 : 00 to 17 : 00 photosynthesis is negative value and there is no photosynthesis accumulation , as well as there is photosynthesis accumulation only during low light intensity on morning or night . Daily change of chlorophyll fluorescence shows that light intensity beyond 800 μmol·m-2·s -1 is not suitable to grow dendrobium , because strong photoinhibition of photosynthesis is occurred before negative
    photosynthesis accumulation when the light intensity is beyond 800 μmol·m-2·s -1 in natural light . The experiments controlling light intensity show that ETR rises rapidly , rate of electronic transmission of photosynthesis and NPQ increase , light energy can be used effectively and surplus light energy can be dissipate at 500μmol·m-2·s -1 light intensity as well as dendrobium has much better responsibility for light .

  • Resistance to birdcherry aphid 〔Rhopalosiphum padi (L. ) 〕in 35 turfgrass varieties from 7 genera in Triticeae was studied. 15 high resistant , 3 middle resistant and 2 low resistant varieties were identified. Both creeping bentgrass and buffalograss have the best resistance and are in immune level . 4 of the 6 bermudagrass varieties are also immune from the pest . Resistance in tall fescue varieties varied from immune to low resistant and Kenturky bluegrass from high resistant to middle susceptible. Zoysiagrass exhibited low or middle susceptible resistance level . Ryegrass varieties , however , are all high susceptible to the pest . The analysis of the symptom and the correlation between aphid density and plant injury degree suggested that antibiosis and tolerance contribute to the resistance in turfgrasses to the insect .

  • There are about 213 species of wild fruit trees in Hubei province , central China , which belong to 26 families and 44 genera. Most of them are Rosaceae and berry fruit trees. They form three concentrated distributing centers : Shennongjia and its surrounding , southwest of Hubei including Yichang district and Enshi , southeast and northeast of Hubei . These wild fruit tree resources have compatibility of fruit trees resources both in south and north China. They also have the characteristics of diversity for deserving exploitatiing and utilizating. However ,there resources are threatened by the alteration ecological environment and humanps unreasonable utilization with natural resources , so the integrative research in conservation , exploitation and utilization should be strengthened.

  • The results of studies on pollinated fruitfulness for apples showed that the self2fruit set percentage for a large number of cultivars , except ‘Megumi’, was low , which represented self-incompatibility.   • Biennial and potted‘Shuangyou’wild grape ( Vitis amurensis Rupr. ) cultivar was used for the trial material and its distribution of assimilation products was studied by 14C tracing technique. The results showed that the 14C assimilation products were mainly distributed to the leaves fromflowering to fruit-coloration stage , the export distribution rate of leaves was 76.69 % - 45.09 %. During fruit ripeness the 14C assimilation products were mainly distributed to the fruit and the export distribution rate of fruit was 47.39 %. It shows that the leave have higher competition ability to absorb assimilation products than fruit during most growth and development stages. The fruit-coloration stage was key stage of assimilation products transition from leaves to fruit . The fruit ssimilation products distribution rate was higher during fruit growth than that of during flowering and fruit ripening stages.

  • Ziziphus jujuba cv. Dongzao , light embryo abortion cultivar and Ziziphus jujuba cv. Lizao , high embryo abortion cultivars were employed to determine endogenous hormone of pulp and seed by using HPLC. The results showed that : content of zeatin (Z) in the pulp of Lizao within 20 days after fruit set was higher than that in the pulp of Dongzao , so did the content of GA3 and IAA within 30 days after fruit set ; ABA in the pulp of Lizao was found earlier than that of Dongzao. In Dongzao , Z content of seed was much higher than pulp which reached peak 20 d after fruit set , GA3 content of Dongzao seed kept low all along. IAA content of Dongzao seed reached peak 40 d after fruit set , and kept high level after that , much higher than pulp of Dongzao ; IAA/ GA3 , IAA/ Z kept low within 55 , 40 days separately after fruit set , but raised very fast after that . Three growth hormones in the pulp of seedless fruit were higher than seeded fruit Z , GA3 and IAA content in the pulp of seedless fruit were higher than seeded fruit in Lizao 10 d after fruit set . The high Z , GA3 and IAA level in pulp were unfavorable for embryo development , but high Z and IAA level in embryo were favorable for embryo development .

  • The experiment was conducted with five species of citrus Huanglongbing material . On the base of the bioassay , we used PCR and Nested2PCR were used to detect precisely. The special DNA fragment was cloned and sequenced. The results showed that : both of these two PCR methods can detect the material with no obvious symptom. However the Nested-PCR was better : It could detect the Catharanthus roseas and Citrus reticulata Blanco just within one or two months after the inoculation ; but when PCR was used , the pathogen could be detected over two months after inoculation. The lowest concentration of DNA that can be detected by Nested2PCR was lower than fg , yet PCR is only pg. The sensitivity of Nested2PCR is nearly ten thousand times higher than that of PCR.

  • The meiotic behavior and male gametes development in cytoplasmic male sterile lines (CMS) 21A and Jin A of hot and sweet pepper ( Capsicum annuum L. ) and their maintainer lines 21B and Jin B were studied in this paper. The results showed that microsporogenesis could process normally until former uni2nucleate pollen stage. There are not differences in the meiosis of both A lines and B lines , and between CMS of hot and sweet pepper. At metaphase I , the chromosome constitution was composed of nine ring bivalents and three rod bivalents. Equal chromosome divisions resulted in two poles with chromosome n=12 at anaphase I , and four poles at anaphaseII. After tetrad stage , uni-nucleate pollen underwent a progressive process of collapse , owing to being squeezed by bnormal swelled tapetal cells. Bi-ucleate pollen could not produce since the second pollen mitosis failed to occur ,and the gametes became completely sterile.

  • pBI1212based vector pZM 612 containing an antisense full2length 2b gene of CMV2BG was constructed and introduced into Nicotiana tabaccum var. SR1 , via Agrobacterium tumef aciens2mediated leaf2disc transformation. The regenerated plants were screened by PCR , and further verified by PCR-Southern and one-step RT-PCR. The results indicated that antisense 2b gene of CMV2BG had been integrated into the tobacco chromosomes ,and expressed at transcriptional level . The resistance of transgenic plants was observed after viral infection , indicating the mediation of resistant to CMV2BGby the antisense 2b gene. The antisense 2b gene of CMV2BG (pZM 612) was also introduced into Lycopersicon escuentum var. Hongmanao 213 , and PCR positive transgenic tomato plants were obtained.

  • Differences of Shelf-life in Some Melons Including Six Varieties of Cucumis melo L.

  • Thirteen melon accessions ( Cucumis melo L. ) including six varieties were used to investigate the difference of shelf-life. Results revealed that five accessions from var. saccharinus and inodorus appeared lower ethylene production rate and weight loss rate , displayed a very good shelf-life. Although var. reticulatus showed a rather low ethylene production rate , their shelf2life was shorter than that of var. inodorus. Var. cantalupensis ,makuwa and acidulus appeared comparatively shorter shelf-life. Moreover , it was observed that weight loss , ethylene production and shelf-life are independent of the nature of rind whether netted or smooth.

  • Chloroplast ultrastructures changes in mesophyll cells in response to heat stress in thermo-resistant (02276) and thermo-sensitive (L402) cultivars of tomato were investigated. Heat stress damaged the regular ultrastructure of the envelopes and thylakoid systemof chloroplasts in mesophyll cells of tomato. There existed significant differences in the effects of heat stress on chloroplast structure between thermo-resistant and thermo-sensitive cultivars. The changes of chloroplast ultrasture under heat stress could be used as an important index to appraise the heat-tolerablity at the cellular level .

  • The occurrence of diseases and changes in activities of the three protective enzymes , phenylalanine ammonialyase (PAL) 、peroxidase ( POD) and superoxide dismutase ( SOD) in leaves of Cymbidium sinense,were studied after inoculating Colletotrichum gloeosporioides. The results showed that the activities of PAL increased significantly after 36 h inoculation with C. gloeosporioide , and reached the highest activities at 84 h. POD activities appeared two peaks at 12 h and 60 h , respectively , after inoculation. The first peak surpassed the second peak. SOD activities rose quickly after inoculation with C. gloeosporioide , and reached the highest level at 12 h ,
    then fell . The increase in the activities of PAL , POD and SOD showed that these three enzymes play an important role in resisting the infection of C. gloeosporioide.

  • ‘Liaozhen 2’, a promising dwarf apple rootstock , was derived from the cross of M. Pruniforlia ×M9.The results of 222year investigation show that its characters of dwarf , precocity , productivity and rooting ability are similar
    to M26. The cold hardiness of ‘Liaozhen 2’is stronger than that of M26. The grafting compatibility with Malus baccata(L. ) , and commercial apple cultivars (Fuji , Ralls , Yueshuai , etc) is good. The trees on‘Liaozhen 2’begin to fruit
    in the third year and the yield will reach 22500 kg/ hm2 in the fifth year.

  • ‘Tainong Tianmi’is a new peach cultivar introduced from Taiwan province. The origin is Brazil named‘Premier’. It can flowering and bearing under the condition of 54 hours of ≤7.2 ℃ during the season of winter. It is a high-yielding variety. The fruit yield was 27.5 t/ hm2 and 38.4 t/ hm2 after planting 2 - 3 year , respectively. The soluble soild content of 10.3 % - 14.5 % and 147.8 g of single fruit .

  • ‘Jinxia’is a tetraploid cultivar derived from the wild kiwifruit population of Actinidia chinensis ,The fruit is sub-cylindrical with maxium 134 g. The fruit setting rate is very high and the quality of fruit is superior with 15 % soluble solids and 1100 mg/ kg amino acid. The fruits could be stored for 30 days in ambient atmo-sphere.

  • ‘Lüdao 1’is a new special cultivar for greenhouse cultivation. Its plant grows moderately. The first florescence locates at the third or fourth joint place of the plant . The length of the fruits is 25 - 30 cm with bright-green surface , and white spines sparse on the fruit skin. Its taste is excellent . Its yield increases about 49.5 % than the control cultivar of‘Changchunmici’. This cultivar is most suitable for the winter and spring protected field in north of China.
