  • In this study,an IDD transcription factor gene PpIDD11 was cloned from peach‘Qiumihong’. The coding sequence of PpIDD11 is 1 575 bp,it encodes 524 amino acids and has a conserved ID domain. The assay of subcellular localization showed that PpIDD11 was localized in the nucleus. PpIDD11 showed the transcriptional activation activity in yeast. Quantitative real-time PCR analysis indicated that PpIDD11 was mainly expressed in floral organ,with a highest transcript level in pistil. Overexpress PpIDD11 Arabidopsis exhibited a phenotype of higher stigma,shorter siliqua and fewer seeds compared with wild type. In addition,the transgenic lines showed a curled rosette leaves and dwarf phenotype. Transcriptome sequencing analysis showed that the PpIDD11 transgenic Arabidopsis line had 3 378 up-regulated genes and 3 156 down-regulated genes,including the regulatory genes(LOX4,LOX3,GLC,E6L1,etc)of floral organ development.

  • To explore the heredity variation and inheritance trend of flower and fruit traits in the F1 progenies of Chinese cherry [Cerasus pseudocerasus(Lindl.)G. Don],17 major traits of 708 individuals derived from true hybrids of two distinct cultivars‘Hongfei’(with large fruit size and excellent comprehensive performance)and‘Pujiang Honghua’(a semi-wild germplasm with small fruit size)were analyzed. Under the climatic conditions of Chengdu City in Sichuan Province,the blooming date of F1 progenies ranged from 7 to 17 February 2021,and mostly between 12 and 14 February. The fruit mature date started from 30 March to 19 April,with high frequency between 5 and 15 April. White,pinkish and pink petal color separation,and orange red,red,purple red and black purple peel color separation occurred in the F1 progenies,showing abundant genetic variation of ornamental phenotypes. Seven important fruit traits,including fruit weight,size,fruit shape index,total soluble solid content and stalk length followed normal distributions,a typical sign of quantitative trait that might be controlled by polygenes. The broad-sense heritability(Hb2)of these traits ranged from 60.12% to 84.90%,and the coefficient of variation(CV)was between 4.77% and 20.75%. The average fruit weight of F1 progenies was 3.71 g(range:1.60-6.37 g),smaller than the mid-parent value,with the Hb2 and CV value being 82.50% and 20.75%,respectively. The ratio of individuals with fruit weight higher than high parent(HH)and mid-parent(HM)was 28.81% and 2.26%. Ellipse,reniform,near round,heart and oblateness shape fruits were observed in the offspring. The former three shapes accounted for 32.34%,29.10% and 21.89%,indicating a tendency toward ellipse and round heredity. Total soluble solid(TSS)content,ranging from 8.38% to 20.53% and the mean value of 13.03%,had over high-parent tendency in F1 progenies,with the Hb2 value of 76.77%. 47.60% and 76.13% of individuals were higher than that of high parent and mid-parent respectively. As for fruit flavor,sweet-sour,sour-sweet and sweet accounted for 90.95% proportion of all hybrids. The fruit stalk length showed heterosis with an HH value of 51.13%,which was beneficial for fruits harvest,storage and transportation,as well as the cultivation of ornamental cultivars. Correlation analysis indicated that the fruit weight was positively correlated with the longitudinal,transverse and lateral diameters(r = 0.880**-0.946**),but negatively correlated to the TSS content(r =-0.103**). These results will provide a basis for the prediction of offspring' main economic traits and parental selection in breeding programs of Chinese cherry.

  • To investigate the expression patterns of miR408 and its target DlLAC12 during longan globular embryo(GE)development and different abiotic stresses. The miR-RACE PCR and Tail-PCR were used to clone the cDNA and gDNA of dlo-miR408 and cDNA of DlLAC12,and the transcriptional start site(TSS),and promoter of MIR408(pro-MIR408)were ascertained. Characteristics of dlo-miR408 sequence,homologous and cis-acting elements of the promoter were bioinformatically analyzed. The transcriptional expression levels of the members of dlo-miR408 family at various abio-stress conditions were verified with qRT-PCR,and the expression patterns of dlo-miR408-3p with its target gene DlLAC12 were further analyzed at early somatic embryogenesis stages with various plant growth regulator treatments. pro-MIR408 sequence contains hormone signal transducing cis-acting elements such as ABA,GA3 and JA,and light and low temperature stress cis-elements. The results showed that the length of pri-miR408 cDNA,gDNA were 706 bp. The TSS was“C”. The length of DlLAC12 cDNA was 1 725 bp. The length of pro-MIR408 was 1 532 bp. dlo-miR408-3p showed dynamic expressions in responding to sucrose,Cu2+ and temperature treatments by means of qRT-PCR,whereas dlo-miR408-5p1 showed downregulation in the imbalanced Cu2+ and low temperature conditions,and maintained stable expression to sucrose changes. Moreover,dlo-miR408-3p and DlLAC12 responded to high concentration of ABA(≥ 500 μmol · L-1)with negative regulations,and GA3 treatment made them same in the expression pattern. The negative regulation at the late globular embryo(GE)developmental stage played an important role in the GE morphogenesis. The present work showed that dlo-miR408 and DlLAC12 were involved in the longan somatic embryogenesis and abiotic stress.

  • To study the regulatory mechanism of the anthocyanin synthesis pathway in pomegranate,we isolated PgMYB111 gene from the petals of‘Liuhuahong’and‘Liuhuabai’. Results showed that the cDNA sequence contained a 1 101 bp open reading frame(ORF)encoding a 366 amino acid polypeptide. Multiple alignment analysis of amino acid sequences showed that the N-terminus of PgMYB111 protein contained a conserved R2R3 domain and a binding site of [D/E]Lx2[R/K]x3Lx6Lx3R interacted with bHLH protein. Furthermore,the amino acid sequence had the highest similarity with MYB transcription factors in Malus × domestica and Prunus dulcis. Subcellular localization assays suggested that PgMYB111 localized in the nucleus. qRT-PCR analysis indicated that the expression of PgMYB111 gene in‘Liuhuahong’ was 4.3 times higher than that in‘Liuhuabai’. In addition,the anthocyanin content in‘Liuhuahong’petal is significantly higher than that in‘Liuhuabai’petal. The results of tobacco transient expression showed that the expression level of the gene increased at first and then decreased. The anthocyanin content in transgenic tobacco leaves was consistent with the gene expression,and reached the highest level on the fifth day. They were 3.8 times and 8.9 times higher than those in uninfected leaves,and 3.17 times and 5.57 times higher than those in pBI121 leaves,respectively. In summary,PgMYB111 could involve in the anthocyanin synthesis pathway and facilitate its synthesis.

  • SmARF5 gene was cloned from the eggplant cultivar‘S12’. The open reading frame is 2 793 bp,and it encodes a 930 aa protein that located in the nucleus. The pGBKT7-SmARF5-Full and pGBKT7-SmARF5-674aa vectors were constructed. The transcriptional activation activity of SmARF5 protein was analyzed and showed by yeast transformation. The full length of SmARF5 could interact with SmIAA16 and SmIAA26 proteins. The truncated SmARF5-674aa did not interact with SmIAA16 or SmIAA26. The pCAMBIA-2301G-SmARF5 overexpression vector was constructed and genetically transformed into tobacco plants. Compared with the wild type,obvious branches and the stem thickness were increased in the transgenic plants. The above results indicated that the SmARF5 transcription factor might play an important role in the formation of plant branches through the auxin signal transduction pathway.

  • Celery seeds are small and contain a large amount of germination-inhibiting substances,which brings difficulties to celery seeding propagation. The seeds of‘Liuhe Huangxinqin’and‘Saixue’were treated with different concentrations(0,5%,10%,20%,40%)of NaClO cultured in petri dish and acupoint dish. The seed germination index,seedling growth index and photosynthetic parameters were measured. In addition,the changes of chlorophyll content and related gene expression in seedlings were analyzed. The results showed that celery seeds treated with 5% NaClO had the highest germination rate,germination potential,germination index and vigor index,and the strongest seedling growth potential. And the seedling vigor index,chlorophyll content and net photosynthetic rate were also increased to some extent. Also,the relative expression levels of related genes were also higher. These results suggested that the appropriate concentration for NaClO to promote celery seeds is 5%.

  • In order to explore the function of alternative oxidase(aox)gene in high temperature stress response of macrofungi,the aox gene was cloned from Pleurotus ostreatus CCMSSC00389 strain. The total length of gDNA and cDNA were 1 845 bp and 1 152 bp respectively,encoding 383 amino acids and two transmembrane helix regions. The molecular weight of the protein was 43.53 kD. The function and regulatory pathway of aox under high temperature stress were verified by wild-type strain and transfor- mation strains of aox gene. The results showed that the mycelial growth rate decreased by 42.27% under 32 ℃ high temperature stress. The increase of aox gene transcription level may up-regulate the expression of antioxidant enzyme gene by mediating retrograde signal pathway,and then promote the clearance of ROS and improve the mycelial growth rate under high temperature stress. The results showed that aox gene played a positive regulatory role in improving high temperature tolerance of P. ostreatus mycelia.

  • In this study,the contents of anthocyanins in the tepals were determined from flower of red Tulipa gesneriana‘Strong Fire’at four developmental stages by high performance liquid chromatography(HPLC). The results showed that the content of anthocyanins continued to increase along with the development of flowers,and the highest content was up to 3 002.75 μg · g-1 FW in fully flowering stage flower. The sequence of TgANS was cloned,and the GenBank accession number was MZ442383. TgANS has 1 071 bp ORF region,encoding 356 amino acids. The expression level of TgANS differed significantly at different flower stages,and showed a trend of rising during the flower development. The expression pattern was consistent with the accumulation of anthocyanin in the four stages of flower development. Ectopic expression of TgANS in transgenic tobacco plants showed that the flowers of transgenic plants obviously turned red and TgANS was expressed at high levels in transgenic plants. The anthocyanin content in the tepals of the transgenic plants was slightly higher than that in empty vector control. Further analysis of the flower color related genes indicated that the expression of genes NtF3H,NtF3’H and NtDFR were increased significantly,and NtCHS,NtCHI were decreased in transgenic tobacco plants. These results indicated that tulip TgANS gene is involved in the biosynthesis of anthocyanin.

  • Cutting of Rosa chinensis‘Old Blush’were used as the material to clone the AP2/ERF transcription factor RCDREB2A gene based on transcriptome under drought stress. The RcDREB2A gene contained a 1 155 bp open reading frame(ORF),encoding 384 amino acids and contained a conserved AP2 domain between 75-142 amino acids. Phylogenetic analysis showed that RcDREB2A has the highest affinity with Fragaria veaca FvDREB2A(XP_004307690.1). RcDREB2A protein has a predicted molecular weight of 5 737,a theoretical isoelectric point of 4.75. RcDREB2A protein has no transmembrane domain and no signal peptide. RcDREB2A protein was localized in the nucleus and showed no transcriptional activation activity. RcDREB2A was highly expressed in leaves,and it could be up-regulated by ABA,drought,low temperature and salt stresses. The above results indicated that RcDREB2A may participate in a variety of abiotic stress responses of rose and plays a crucial role in resisting abiotic stresses.

  • An R2R3 MYB gene,named RhMYB113c,was cloned from cDNA of the petal in Rosa hybrida‘Carola’. The open reading frame of RhMYB113c was 699 base pair,encoding 233 amino acids. The multiple sequence alignment analysis showed that RhMYB113c contained R2 and R3 domains. Phylogenetic tree results indicated that RhMYB113c was closely related to anthocyanin synthesis activator AtMYB114,AtMYB113,AtMYB75,and AtMYB90. Tissue specific expression showed that RhMYB113c was mainly expressed in petals and styles in‘Carola’,but not in all tissues of white flower variety ‘White Satin’. Gene expression at different flower development stages indicated that RhMYB113c was not expressed in‘Carola’uncolored petals,highly expressed in full flowering stage,but not in all stage of white flower varieties‘White Satin’. Yeast two hybrid and tobacco transient expression experiments suggested that RhMYB113c may regulate anthocyanin synthesis and determine the color of rose petals.

  • The tea plants(Camellia sinensis)have a strong Se-enriching capacity,however,the physiological characteristics of Se uptake are still unclear. In this study,the effects of solution pH,cultivation time,Se concentrations and the energy metabolism/aquaporin/anion-channel inhibitors on selenate and selenite uptake in the roots of ‘Zhongcha 108’were investigated. The results of pH treatments found that the uptake rate of selenite decreased with the increase of pH. The uptake rate of selenate reached the highest at pH 3.0,and remained stable from pH 4.0 to pH 7.0,with no significant difference. Time-course analysis showed that there was a cumulative effect on the uptake of selenite and selenate by tea plant roots. The total Se content in roots increased significantly with the increase of cultivation time under selenite treatment,while showed a trend of increase first and then decreased under selenate treatment. Under different Se concentration treatments,there was a linear positive correlation between the uptake rate of Se and the solution Se concentration. The uptake rate of selenite was significantly higher than that of selenate. To investigate the pathways via which the selenate and selenite uptake of tea plant root,inhibitors treatments were carried out under pH 5.0 and 5 μmol · L-1 Se conditions. The results showed that both of selenate and selenite uptake of tea plant roots were inhibited significantly by the energy metabolism inhibitors of CCCP and DNP and the aquaporin inhibitor of AgNO3,indicating that the uptake of selenate and selenite were energy-dependent and aquaporins-mediated processes. The uptake rate of selenite was inhibited by NPPB,however,the uptake rate of selenate was not affected by NPPB,indicating that the anion-channel was also the pathway for selenite uptake in tea plant root.

  • The wild apple tree Malus sieversii(Rosaceae)is considered to be the ancestor of modern cultivated apple trees. To effectively conserve apple genetic resources,it would be important to elucidate differences of chloroplast genomes between M. sieversii and cultivated apples,as well as verify whether modern cultivated apples originated from M. sieversii. In this study,we used the next-generation sequencing technology to complete the whole-genome sequencing,assembly,and annotation of M. sieversii as well as two domesticated apple trees‘Red Delicious’and‘Golden Delicious’. We successfully mapped the chloroplast genomes of M. sieversii and the two domesticated apple trees,and subsequently constructed the Malus phylogenetic tree at the level of the chloroplast genome. Through a series of comparative analyses,We obtained the following main results:the chloroplast genome of M. sieversii exhibites a typical quadripartite structure,and the complete genomes ranged between 160 068 and 160 288 bp in length. A total of 131 genes were annotated in M. sieversii and the two domesticated apple trees,including 86 protein-coding,37 tRNA,and eight rRNA genes. The base composition of the chloroplast genomes of M. sieversii and the two domesticated apples varieties were similar. A total of 43,49,and 43 repetitive sequence and 64,61 and 57 simple sequence repeat sites were identified in M. sieversii,‘Red Delicious’and‘Golden Delicious’,respectively. For the simple sequence repeats,the main repeat types were single A or T base repeats. Finally,phylogeny study showed that all the Malus species formed a single monophyletic group(BP = 100). M. sieversii and the two cultivated varieties were grouped in a subclade with strong support(BP = 100),supporting the hypothesis that M. sieversii is the putative ancestor of domesticated apples.

  • The banana cultivar‘Fenza 1’is the only Fenjiao(Musa spp. ABB,Pisang awak)cultivar with resistance to Fusarium wilt in the current commercial banana plantings. The cultivar came from the seeds seedling obtained accidently. Its genomic composition and male parent are unknowen. In this study,Flow cytometry(FCM),chromosome flaking,molecular marker technology ITS-PCR-RFLP and copia-IRAP,cooperate with characters taxonomic scorecard were used to identify the genomic composition of‘Fenza 1’. The ploidy was tetraploid showed by the FCM. The chromosome number was 2n = 4x = 44 showed by the chromosome flaking;its genomic composition include A and B showed by the ITS-PCR-RFLP;two or more than two B genomic groups showed by the copia-IRAP;the score of the characters taxonomic is 67,and the corresponding genomic composition is ABBB. Based on the results above,we prosposed that the genomic composition of‘Fenza1’is ABBB,and speculated its male parent may be wild banana with the genomic composition BB.

  • Identification and Sequence Analysis of Tomato Spotted Wilt Orthotospovirus Infecting Lettuce in Sichuan

  • Biological inoculation,electron microscopy and small RNA sequencing were employed to identify the virus infecting lettuce in Pengzhou,Sichuan Province,followed by reverse transcription(RT)-PCR for verification. Mechanical inoculation showed similar symptom on back inoculated lettuce to field samples,and necrosis was observed on Petunia hybrida leaves. Numerous virions which had similar morphology characteristics to members of the genus Orthotospovirus were observed,and they clustered in the cisternae of endoplasmic reticulum in the mesophyll cells. Nearly complete sequence of tomato spotted wilt orthotospovirus(TSWV)was obtained by small RNA sequencing,revealing that the pathogen in infected samples might be TSWV. RT-PCR results indicated the presence of TSWV in both field and inoculated lettuce plants and indicator plant,further conformed that TSWV was the pathogen causing lettuce virus disease in Pengzhou. This was the first report on TSWV infecting lettuce in Sichuan. The phylogenetic tree constructed based on assemble sequences showed that Pengzhou isolate(TSWV-SCPZ)had close relationship with Korea isolate (YY and K),whereas more close related to America isolate(MR-01)based on N gene. These isolates shared a similar evolutionary pattern with other isolates present in Korea and America.

  • Diversity Analysis of Melon Fruit Color Based on the Lab System

  • The genetic diversity of eight qualitative traits and 11 quantitative traits(17 indicators)of 466 melon germplasm materials were analyzed,which were selected from collected and preserved resources by our research group. For the quantitative analysis of fruit color,the La and b values of the peel,the outer flesh and inner flesh were determined by precision colorimeter. The results showed that the diversity indexes of three quality traits and 11 quantitative traits(17 indexes)were higher than 1,which indicated that the 466 selected melon materials used in this experiment had high genetic diversity. In this study,melon fruits color were measured digitally by Lab values and color classification were carried out by Lab values,which overcame the disadvantage of inaccurate color description and realized the quantitative analysis of color. Finally this analysis lays the foundation of digital description of fruit color in melon breeding.

  • ‘Meiyu’is a new mid-early ripening pear cultivar derived from the cross of‘Gold Nijisseik’and‘Jinhua 4’. The trees have moderate growth vigor,upright posture,strong budding force and branching ability. The fruit is oblate and round,with an average single fruit weight of 252 g. The peel is yellow green,thin. The fruit stalk is short. The flesh is milky white,crisp,juicy,sweet. The soluble solids content is 12.5%-14.3%,and the titratable acid content is 0.2%. The fruit hardness is 5.7 kg · cm-2. The fruit ripens in early August in Shijiazhuang area,and the fruit development period is 137-142 d. The yield is 45 111.4 kg · hm-2.

  • ‘Yuetong’is a new pineapple cultivar selected by artificial hybrid breeding. The color of leaf is green. There are no spines around the leaf margin. The leaf-tip is soft. There are a few white wax powder stripes on the leaf abaxial surface. The fruit is cylindric. The color of fruit skin is purple-red when ripened,and the color of fruit pulp is yellow-orange with slight purple-red. The flesh is sweet with abundant juice and dietary fiber. It was observed that‘Yuetong’performed well with high and stable yield(58 000 - 72 000 kg · hm-2). The fruit are suitable for table or for processing.

  • ‘GuiJinfeng’ is an early-maturing and medium sized fruit watermelon cultivar. The plant is vigorous,with good fruit setting. The fruit is oblong oval-shaped,and the single fruit weight is 2.5-4.0 kg. The rind is golden. The peel is about 0.6-1.0 cm in thickness. The red fresh is crisp,with SSC of 12.0% in center. The whole growth period is 80-100 d in spring and 60-85 d in autumn. The average fruit development period is 25-30 d. The cultivar has broad adaptability and good perspective of commercialization in Guangxi Province. It is suitable for protected and open field cultivation.
