  • Regeneration and Transformation in Vitro of the Strawberry Varieties

  • An efficiency and stable regeneration system in vitro and genetic transformation system for the commercial strawberry varieties were developed , and transgenic plants were obtained. Bud regeneration percentage of leaf pieces of strawberry varieties ‘Tudla’and ‘Senga Sengana’reached 100 %. Leaf pieces from in vitrogrown plants‘Tudla’were cocultivated for 3 days with Agrobacterium tumef aciens EHA105. After the cocultivated leaf pieces were incubated on regeneration medium
    containing 40 mg/ L kanamycin for 4 weeks , the transformed buds appeared on the explants. The regeneration percentage of transformation buds of strawberry variety‘Tudla’reached 6. 8 %. The result of histochemical staining of GUS gene showed , of the 10 independent plants transformed with GUS gene , 5 expressed GUS activity strongly. The plants transformed with bar gene were able to differentiation on the medium containing glufosinate 10 mg/ L , and expressed strongly resistance to glufosinate
    on field. The transgenic plants flowered and fruited normally.

  • The parthenogenic young embryos were gained in apple varieties (Bela , Liaofu and Fuji) due to pollination with radiated pollen by 1 000 - 1 500 Gyγ ray. The haploids of these varieties were produced by young embryo culture and regeneration. The investigation indicated that major factors which limited hapliod production efficiency in apple by such technique included abscission of
    fruits in early stage , young embryo abortion and difficulty in regeneration of young embryo with endosperm. The results also suggested that regeneration of 25 d young embryo (ovule) was available.

  • The calcium contents in all parts of bitter pit apples were significantly lower than that of the sound ones , but only the contents of magnesium at the outer cortex significantly higher than the sound ones. The activities of superoxide dismutase (SOD) , peroxidase ( POX) , catalase (CAT)
    and ascorbate specific peroxidase (APX) in the pitted fruits were significantly lower than that of the sound. Vacuum infiltration of apples with CaCl2 100 mmol/ L increased activities of the above antioxi dant enzymes , and the treatment of MgCl2 decreased the activities of POX, APX and CAT , implying
    the activities of these three enzymes were regulated by calcium. The contents of malonaldehyde and the relative conductivity were higher in bitter pit apple fruits , which showed that the peroxidation of membrane might be the important reason of the physiological disorder in apple with lower level of calcium.

  • A peach cultivar‘Hujingmilu’was used to investigate the changes of LOX activity , ethylene production and memberane fatty acid composition. It was found that the major fatty acids in the ripening peach fruit included palmitic acid (16∶0) , stearic acid (18∶0) , oleic acid (18∶1) , linoleic acid (18∶2) and linolenic acid (18∶3) . There were remarkable changes in the proportion of
    oleic acid , linoleic acid and linolenic acid , especially linoleic acid and linolenic acid , while no such notable changes in the proportion of palmitic acid and stearic acid during fruit ripening at 20 ℃. Linolenic acid was mainly used as a LOX catalyzing substrate before LOX self-propagation during fruit
    ripening at 20 ℃. LOX might play a role in memberane lipid peroxidation which participated in fruit ripening and softening. The significance of these results was discussed.

  • China possesses more than 70 % ginkgo resources in the world. The primary problems existing in ginkgo cultivation are : (1) no big-grain and fine nucleus-used cultivars ; (2) no known genetic law of primary characters ginkgo seed. The purposes of this research are to collect 50 nucleusused cultivars from 12 provinces in China , to carry out genetic analysis for economical character and
    multiple character selection on ginkgo seed. The results of analysis show that maxmum single nucleus weight , stone mass/ fruit mass ( %) and kernel mass/ stone mass ( %) are 3. 89 g , 33. 58 % and 82. 49 % , respectively. The heritability ( h2) of single nucleus mass is 0. 9759. The genetic correlation coefficient ( rG) between nucleus2shape coefficient and single nucleus mass , and direct genetic path coefficient ( PY·i) of single fruit mass to single stone mass are 0. 9986 and 1. 407 , respectively. Twelve provenances from China are divided into four classes. Three fine nucleus-used cultivars are selected out through index or direct selection methods from fifty nucleus-used cultivars in China. The theory of nucleus2used cultivar selection was studied in this paper.

  • Under the stress of low temperature (15 ℃) and low light intensity (100μE·m-2·s-1) , the photosynthetic light compensation point of cucumber seedlings went up at the beginning of treatment , some parameters of chlorophyll a fluorescence such as Fv/ Fm , qp , ΦPS Ⅱ ( Y) decreased obviously , and the activity of RuBPCase was also reduced. This results indicated that the primary light energy conversion of PS Ⅱ, electron transport and carbon assimilation of cucumber seedlings had been inhibited , thus the capacity of its utilizing low light intensity went down. However , after
    the induction of stress , the adaptability of cucumber seedlings increased , and the above parameter reversed the level before stress treatment . The results also showed that the adaptability of greenhouse varieties was higher than that of open field varieties.

  • The tests of daily variation of photosynthetic rate of butterhead lettuce grown in green house are conducted. The results show that the daily variation of photosynthetic rate is a two peak curve in sunny days in summer and autumn season. While in winter with lower temperature, it is a
    one-peak curve pattern. In the cloudy and raining days, the change of photosynthesis just follow the change of radiat ion. The photosynthesis maintaining at its high value is not more than 3- 4 hours within a day. If the radiation and temperature cannot meet the need of crop in the morning, it is still possible to produce a photosynthet ic peak in the afternoon with suitable environment factors for the crop photosynthesis.

  • The sweet protein gene MBL II was introduced into lettuce by co􀀁culturing excised cotyledon explants with Agrobacterium tumefaciens . The Agrobacterium strain LBA4404 contained the binary vector pLX10, which carried sweet protein gene MBL II, select ive gene NPTII and reporter
    gene GUS . The best transformation efficiency was obtained when explants were co􀀁cultured with Agrobacterium for 3 days and cultured on the selective medium containing 50 mg/ L kanamycine ( shoots regeneration rate 58. 3%, albino rate 29% ) . Roots were induced in all adventitious shoots
    on MSII (MS+ NAA 0. 05 mg/ L+ Kan 50 mg/ L+ carbencillin 300 mg/L) . The transgenic nature of regenerants were demonstrated by plant growth on medium containing kanamycine, GUS histochemical assay and PCR amplification. 7. 6% of all Kanamycine resistant shoots was GUS positive.
    Southern blot results confirmed that the MBL II gene had been incorporated into the genome of lettuce.

  • The studies of the formation and development of the sexual organs on the primary and secondary flower cluster of 􀀂Kyoho grapevine were conducted, aiming at elucidating the mechanism of the low seed production on the
    primary flower clusters. The results showed that the pollen of the florets of the secondary flower clusters was smaller, though the percentage of pollen stained by acetocarmine on florets of both flower clusters was same, the percentage of germination on agar of the secondary flower clusers was higher than that of the primary flower clusters. The result indicated that the pollen quality was not related to flower shatter. The embryo-sac evelopment with function cell in the pistils of the secondary flower clusters was more favorable than that of the primary flower clusters. At anthesis, a large number of abnormal embryo􀀁sac were observed in the ovaries of the primary flower clusters. As a result, at anthesis, a large number of abnormal embryo􀀁sac may suppress fertilization and lead to lower seed.

  • Peroxidase isoenzymes of leaves in Ziz ip hus spinosus Hu. and 17 varieties of Z. jujuba Mill. were analyzed by means of perpendicular􀀁plate type of polyacrylamide gel electrophoresis. Fuzzy cluster analysis was conduc􀀁
    ted based on the similarity coefficients of the isoenzyme zymogram, using the R􀀁chain link method of similarity relation ship. Z . spinosus Hu. and 17 varieties of Z. jujuba Mill. were classified into several groups. and the relationship among the groups was discussed. From the phenotype of molecular enzyme level, it has been proved that Z. spinosus is the primeval species of the cultivated Z. jujuba, which is the result of a longtime evolution. But both species are closely related to each other.

  • Basing on the research advancement of taxonomy for Malus trilobata Schneid. , using the theories and methods of phylogeny􀀁phytogeography, the authors found that Malus trilobata developed independently from Malus laoensis Chev. , one of the most primitive species of Malus Miller, after they separated from a homogeneous flora in early time. Malus trilobata is one of the most closely related taxa of the genus Malus Miller and it should be originated in theCretaceous period. As a conclusion, the author suggested that Malus trilobata Schneid. be named Cormus Decisne.

  • The female parent 9704A of ‘Xiangyan No. 14’is male sterility line bred from a sterile plant found in ox2horn pepper No. 21 and the male parent 5904 is an ox-horn pepper inbred line selected from a landrace in Guangxi ,
    with large fruit and thick wall. The late pungent F1 hybrid is suitable for upper and middle region of Yangze river as trans-summer production and South China as winter open field production. The ox-horn shaped and straight fruit with smooth appearance and thick skin is suitable for storage and transportation. It can be harvested both light green (ripening) and bright red (ripened) . The average yield is 49. 5 - 60 t/ hm2 with fruit weight proximately 38 grams.

  • Shannong No. 5 is a new cultivar for protected cultivation. It is suitable for solar greenhouse production in Huabei district. Its plant grows vigorously, and it yields 120 tons per hm2 . Average fruit is about 35 cm length and 150 g weight. Fruits are green, sweet and crispy. It is tolerant to chilling and low light. It is resistant to fusarium wilt, downy mildew, powdery mildew and scab.
