苹果LysM类受体激酶基因家族鉴定与表达分析

摘要:

以LysM(PF01476)和Pkinase(PF00069)保守域全蛋白序列为种子序列,在全基因组范围比对分析了苹果(Malus × domestica Borkh.)LysM-RLK家族的成员,对其家族成员的理化性质、进化关系、染色体分布、基因结构等进行了分析;基于已公布的转录组学数据,分析了LysM-RLK基因在苹果组织、发育过程和生物胁迫中的表达情况。通过分析,共获得12个苹果LysM-RLK基因,其氨基酸序列大小介于553 ~ 1 053,分子量介于62.65 ~ 119.04 kD,等电点介于5.11 ~ 7.45,主要位于质膜;根据进化分析将其分为3个亚组,亚组内各成员的外显子—内含子结构呈现较为相似的特征。基因表达数据显示,苹果的4个LysM-RLK基因表达存在较大的组织特异性,7个基因在苹果腐烂病和再植病发病过程中为差异表达。

Abstract:

Conserved protein sequence of LysM(PF01476)and Pkinase(PF00069)domain were used to search the LysM-RLKs from apple(Malus × domestica Borkh.)protein database. The physical and chemical characteristics,evolutionary relationships,chromosomal distribution and gene structure members were analyzed for all of the LysM-RLKs. Additionally,expressions of LysM-RLKs were assayed in specific tissue and development process of apple as well as under biotic stresses based on the published RNA-seq data. In total,12 LysM-RLK genes were identified in the apple genome. The size(amino acid,aa),molecular weight and isoelectric point of LysM-RLKs in apple varied from 553 to 1 053,from 62.65 to 119.04 kD and from 5.11 to 7.45,respectively. The genes mainly sublocalized on plasma membrane. According to phylogenetic analysis,LysM-RLK of apple could be divided into 3 sub-groups. Within a sub-group,LysM-RLKs exhibited similar features of exon-intron structure. Gene expression analysis showed that 4 LysM-RLKs were expressed specifically in different tissue. In addition,expressions of 7 LysM-RLKs were significantly changed when Apple Valsa canker and Apple replant disease occurred. The study contributed to further investigate the functions of LysM-RLKs and disease resistance research in apple.
