梅遗传多样性的SRAP分析

摘要: 为探明梅种质资源间的亲缘关系,利用SRAP标记对梅135份样品进行了遗传多样性分析。17个引物组合共扩增出124个位点,其中多态性位点为106个,多态位点比率87.5%。每个引物组合扩增位点数在4 ~ 12个之间,平均每个引物组合扩增位点数为7.3;通过NTSYS软件计算得到的样品间SM相似系数介于0.556 ~ 0.958,平均值为0.733,显示梅资源间存在一定的遗传差异;根据SRAP扩增结果,利用UPGMA法构建树状聚类图,聚类分析将135份样品分为7组,聚类分析结果与梅种的形态分类系统基本相符。

Abstract: In order to evaluate the genetic relationship among germplasm of Prunus mume Sieb. et Zucc.,sequence related amplified polymorphism(SRAP)marker was used to analyze genetic diversity of 135 samples of Mei. Seventeen primer pairs generated 124 bands,106 bands(87.5%)of which were polymorphic ones. SRAP primer pairs amplified 4 to 12 bands with an average of 7.3 bands per primer pair. Revealed by NTSYS software,the SM coefficient of genetic similarity ranged from 0.556 to 0.958,with an average of 0.733,which indicated that genetic basis was relatively abundance among the 135 Mei resources. A dendrogram was constructed based on SRAP data using UPGMA cluster method. The 135 samples of Mei were classified into 7 major groups,which was basically corresponded with the genetic relationships based on morphological traits.
